A Multiple-Testing Procedure for High-Dimensional Mediation Hypotheses

James Y. Dai et al.

Summary: Mediation analysis is gaining attention in epidemiologic studies and clinical trials, but existing methods have limitations. In this article, a multiple-testing procedure is proposed to accurately control error rates and improve power. Two data analysis examples demonstrate the effectiveness of the proposed method, and an R package is provided for implementation.

On optimal two-stage testing of multiple mediators

Vera Djordjilovic et al.

Summary: Mediation analysis in high-dimensional settings often involves identifying potential mediators among a large number of measured variables. In this work, the impact of threshold selection in screening variables is investigated. A power maximizing threshold is derived and compared with an adaptive threshold. The proposed procedures are illustrated through a case-control study and applied in replicability analysis.

Sparse latent factor regression models for genome-wide and epigenome-wide association studies

Basile Jumentier et al.

Summary: This study introduces sparse latent factor regression models to address statistical challenges in genomic and epigenomic data. Simulation results show that sparse latent factor regression models generally outperform other sparse methods in terms of statistical performance. These models are also more robust to departure from the model in simulations based on empirical data, and they facilitate the estimation of non-null effect sizes and overcome multiple testing issues.

Pregnancy exposure to synthetic phenols and placental DNA methylation - An epigenome-wide association study in male infants from the EDEN cohort

Paulina Jedynak et al.

Summary: In utero exposure to synthetic phenols may lead to alterations in placental DNA methylation, with significant DMRs identified in genes related to important cellular functions and imprinted genes. Placental cell heterogeneity may mediate the effects of exposure on DNA methylation, suggesting potential mechanisms for how certain exposures can impact fetal development.

DNA methylation mediates the effect of maternal smoking on offspring birthweight: a birth cohort study of multi-ethnic US mother-newborn pairs

Summary: This study in a predominantly urban, low-income multi-ethnic birth cohort in the US found that maternal smoking significantly affects newborn birthweight, and fetal DNA methylation may play an important mediating role in the impact of maternal smoking on newborn birthweight.

Epigenetic Alterations of Maternal Tobacco Smoking during Pregnancy: A Narrative Review

Aurelie Nakamura et al.

Summary: In utero exposure to maternal tobacco smoking can lead to epigenetic changes affecting the birth and development of infants, with DNA methylation being the most extensively studied mechanism. Some studies suggest reversibility in certain genomic regions of DNAm and memory of smoking exposure in other regions, especially in cases of smoking cessation before or during pregnancy.

Placental DNA methylation signatures of maternal smoking during pregnancy and potential impacts on fetal growth

Todd M. Everson et al.

Summary: Maternal smoking during pregnancy is associated with disrupted placental functions and poor birth outcomes, as reflected in the placental epigenome. The study identified specific CpGs related to maternal smoking during pregnancy, with implications for environmental response genes, growth-factor signaling, and inflammation in placental function. The research highlights the links between placental DNA methylation, maternal smoking during pregnancy, and poor birth outcomes, providing insight into the mechanisms by which smoking affects placental function and fetal growth.

Statistical methods for mediation analysis in the era of high-throughput genomics: Current successes and future challenges

Ping Zeng et al.

Summary: Mediation analysis investigates the intermediate mechanism by which an exposure influences the outcome. It is increasingly popular in high-throughput genomics studies, but faces challenges in genomics due to the large number of potential mediators and the composite null nature of the mediation effect hypothesis. New high-dimensional mediation methods have been developed to address these challenges, providing guidance for statisticians and computational biologists.
