KDM1A inactivation causes hereditary food-dependent Cushing syndrome

Anna Vaczlavik et al.

Summary: This study identified KDM1A gene inactivation as the cause of food-dependent Cushing syndrome (FDCS) in 90% of primary bilateral macronodular adrenal hyperplasia (PBMAH) cases. Through multiomics analysis of patient tissues, the study classified patients into different molecular groups and suggested genetic screening for ARMC5 and KDM1A for earlier diagnosis and improved management.

Overview of the 2022 WHO Classification of Adrenal Cortical Tumors

Ozgur Mete et al.

Summary: The new WHO classification of adrenal cortical proliferations reflects translational advances in endocrine pathology, oncology, and molecular biology. It highlights the understanding of adrenal cortex pathologies through histological features, ancillary studies, and genetic findings, leading to advances in knowledge of these conditions. The classification redefines the spectrum of adrenal cortical nodular disease, emphasizing the importance of accurate assessment of tumor proliferation rate and diagnostic biomarkers in the workup of adrenal cortical neoplasms.

An Overview of the Heterogeneous Causes of Cushing's Syndrome due to Primary Macronodular Adrenal Hyperplasia (PMAH)

Helaine Laiz Silva Charchar et al.

Summary: Primary macronodular adrenal hyperplasia (PMAH) is a rare cause of adrenal Cushing’s syndrome, and recent advances have revealed the involvement of different molecular mechanisms in its development. The ARMC5 gene is believed to be a major genetic cause of PMAH, while the KDM1A gene is specifically associated with food-dependent Cushing’s syndrome in PMAH.

Genomic classification of benign adrenocortical lesions

Simon Faillot et al.

Summary: This study proposed a pangenomic characterization of benign adrenocortical tumors, identifying four distinct molecular categories and revealing associations between epigenetic alterations and steroidogenesis. The research also uncovered the subclonal nature of chromosomal alterations and somatic mutations in these tumors.

What Did We Learn from the Molecular Biology of Adrenal Cortical Neoplasia? From Histopathology to Translational Genomics

C. Christofer Juhlin et al.

Summary: The incidence of adrenal cortical nodules in the general population is increasing, with complex symptomatology and specific considerations making their diagnosis challenging. Recent advancements in understanding the tumorigenesis of adrenal cortical tumors, including the application of molecular genetics and translational biomarkers, have greatly improved diagnostic capabilities. Comprehensive analyses combining histology, immunohistochemistry, and next-generation multi-omic analyses are necessary to properly distinguish malignancy in some cases.
