Oxidized GAPDH transfers S-glutathionylation to a nuclear protein Sirtuin-1 leading to apoptosis

Syed Husain Mustafa Rizvi et al.

Summary: The study showed that exposure to hydrogen peroxide induced S-glutathionylation and nuclear translocation of GAPDH in HEK 293T cells. Overexpression of Glrx or a redox dead mutant GAPDH inhibited S-glutathionylation and nuclear translocation. Nuclear GAPDH formed a complex with SirT1, transferring S-glutathionylation to SirT1 and inhibiting its deacetylase activity to initiate apoptotic signaling.

Role of Reductive versus Oxidative Stress in Tumor Progression and Anticancer Drug Resistance

Kyung-Soo Chun et al.

Summary: Redox homeostasis is crucial for normal physiological functions and plays a significant role in cancer cell growth, survival, and therapy resistance. Nrf2 is a key stress-responsive transcription factor responsible for maintaining cellular redox balance. Dysregulation of Nrf2 can lead to reductive stress, affecting the development and resistance of cancer cells.

Transglutaminase 2 promotes tumorigenicity of colon cancer cells by inactivation of the tumor suppressor p53

Patrizia Malkomes et al.

Summary: TGM2 plays a crucial role in colorectal cancer, with its transamidation activity being essential for promoting cell survival. By binding to the tumor suppressor gene p53, TGM2 inactivates it, affecting apoptosis induction in cancer cells.

ent-Kaurane diterpenoids induce apoptosis and ferroptosis through targeting redox resetting to overcome cisplatin resistance

Yong Sun et al.

Summary: Ent-kaurane diterpenoids can disrupt redox homeostasis to promote cancer cell death, with compound 23 showing strong inhibitory activity against various cancer cell lines by inducing apoptosis and ferroptosis through increased cellular ROS levels. Compound 23 also sensitizes cisplatin-resistant cancer cells by dual inhibition of Prdx I/II and GSH, suggesting its potential application in cancer chemotherapy.

Cyst(e)ine in nutrition formulation promotes colon cancer growth and chemoresistance by activating mTORC1 and scavenging ROS

Jiao Wu et al.

Summary: Colorectal cancer patients who receive cysteine as part of parenteral nutrition have shorter overall survival, as cystine promotes cancer cell growth by inhibiting certain pathways and conferring resistance to chemotherapy. Manipulating cyst(e)ine content in nutritional formulations may improve colorectal cancer patient survival.
