KDM6B interacts with TFDP1 to activate P53 signaling in regulating mouse palatogenesis

Tingwei Guo et al.

Summary: Epigenetic regulation plays a crucial role in diseases and development. This study focuses on the understudied question of how epigenetic changes affect tissue-specific responses during neural crest fate determination and differentiation. Using palatogenesis as a model, the researchers demonstrate the functional significance of the epigenetic regulator KDM6B in regulating cranial neural crest development and its role in the disruption of P53 pathway-mediated activity. The study also reveals the antagonistic control of H3K27me3 on the Trp53 promoter by KDM6B and Ezh2 in neural crest cells. Furthermore, the researchers show that KDM6B specifically interacts with TFDP1 to activate Trp53 expression in palatal mesenchymal cells. These findings provide mechanistic insights into the epigenetic regulatory network during organogenesis.

Summary: The study found that the level of KDM5B increases in cardiovascular diseases and identified TK-129 as a potent inhibitor with significant effects on activated cardiac fibroblasts. The protective effects of TK-129 are associated with its KDM5B inhibition and blocking the Wnt pathway activation.

The Dual Function of KDM5C in Both Gene Transcriptional Activation and Repression Promotes Breast Cancer Cell Growth and Tumorigenesis

Hai-feng Shen et al.

Summary: Research has shown that the epigenetic regulator KDM5C can both activate estrogen receptor alpha (ER alpha) target genes and suppress type I interferons in breast cancer cells, contributing to cell growth and tumorigenesis. This dual activator and repressor function of KDM5C plays a role in cancer development.

Enhanced Properties of a Benzimidazole Benzylpyrazole Lysine Demethylase Inhibitor: Mechanism-of-Action, Binding Site Analysis, and Activity in Cellular Models of Prostate Cancer

David M. Carter et al.

Summary: A benzyl-substituted variant of an inhibitor targeting KDM4 enzymes demonstrates improved potency, cell-permeability, and cytotoxicity against prostate cancer cells. The inhibition mechanism involves competition with Fe2+ at the active site and binding to a distal site on the enzyme surface. The inhibitor's cytostatic effect is attributed to direct intracellular inhibition of KDM4 enzymes, leading to altered gene expression and epigenetic changes in PCa cells.
