Identification of an ERN1 target site within EGFP mRNA

Marius W. Baeken et al.

Summary: EGFP is commonly used in life sciences research, but this study found that ERN1 targets a specific RNA motif in EGFP mRNA. Introducing a silent mutation in EGFP mRNA prevented ERN1-dependent mRNA decay. Therefore, caution should be exercised when interpreting results using EGFP as a reporter gene in studies involving UPR pathway upregulation, and a mutant lacking the ERN1 target motif may be more suitable for such research.

mRNA-carrying lipid nanoparticles that induce lysosomal rupture activate NLRP3 inflammasome and reduce mRNA transfection efficiency

James Forster et al.

Summary: In recent years, significant progress has been made in the engineering of mRNA lipid nanoparticle vaccines, but there is still limited understanding of their immunogenicity. A design-driven approach is needed to determine the biophysical characteristics that drive nanoparticle immunogenicity. This study investigated the activation of NLRP3 inflammasomes by different lipid compositions in mRNA lipid nanoparticles and found that specific combinations of lipid types significantly influenced their activation.

Summary: The study reports a method that can predict the pKa of lipid nanoparticles (LNPs) from the structure of the ionizable lipid, and investigates the delivery efficiency and factors influencing LNPs both in vitro and in vivo. It was also found that LNPs with more negative charge lead to higher off-target systemic mRNA expression.
