Levosulpiride-loaded nanostructured lipid carriers for brain delivery with antipsychotic and antidepressant effects

Summaira Maqsood et al.

Summary: This study investigated the potential of levosulpiride-loaded nanostructured lipid carriers (LEVO-NLCs) for effective brain delivery with anti-psychotic and antidepressant effects. The optimized LEVO-NLCs were found to have suitable properties for brain delivery, exhibiting sustained drug release behavior and improved pharmacokinetics in the brain compared to the drug suspension. Additionally, LEVO-NLCs demonstrated enhanced anti-psychotic activity and reduced neuroinflammation in a mouse model of psychosis induced by LPS. These findings suggest that NLCs could be effective carriers for delivering active pharmaceutical agents to the brain.

A merged molecular representation deep learning method for blood-brain barrier permeability prediction

Qiang Tang et al.

Summary: The ability to predict the blood-brain barrier permeability of candidate compounds is crucial for central nervous system drug development. Researchers have developed a deep learning-based multi-model framework, Deep-B-3, which uses three types of features to predict the permeability. Validation results using an independent dataset show that Deep-B-3 outperforms existing models, making it a promising tool for drug development.

ApoE-Targeting Increases the Transfer of Solid Lipid Nanoparticles with Donepezil Cargo across a Culture Model of the Blood-Brain Barrier

Gizem Ruya Topal et al.

Summary: Targeting ligands on the surface of nanoparticles can improve the delivery of drugs across the blood-brain barrier. In this study, solid lipid nanoparticles labeled with Rhodamine B and loaded with donepezil were targeted with apolipoprotein E to enhance uptake in brain cells. The potential of ApoE as a targeting ligand for CNS drug delivery was demonstrated through increased uptake and permeability across a BBB co-culture model.
