EZH2 Inhibitors: The Unpacking Revolution

Vera Adema et al.

Summary: This study demonstrates that pretreatment with EZH2 inhibitors can open up H3K27me3-marked chromatin in AML cells, leading to enhanced DNA damage and apoptosis induced by chemotherapeutic agents. The combination of EZH2 inhibitor and doxorubicin also enables the expression of proapoptotic genes, potentially contributing to the death of AML cells.

SALL4 Oncogenic Function in Cancers: Mechanisms and Therapeutic Relevance

Boshu Sun et al.

Summary: This review discusses the overexpression of SALL4 in malignant cancers and its role in promoting proliferation, invasion, and migration. Additionally, SALL4 regulates tumor progression through activation of signaling pathways, gene expression, and affecting the immune microenvironment. Understanding the functions and mechanisms of SALL4 in cancer may lead to novel strategies for cancer therapy.

Demethylation and Up-Regulation of an Oncogene after Hypomethylating Therapy

Yao-Chung Liu et al.

Summary: This study found that hypomethylating agents can induce demethylation and up-regulation of the oncogene SALL4 in certain cases, which is associated with poorer prognosis. The critical CpG island for SALL4 expression was identified in the 5' untranslated region. This discovery highlights the need for further investigation into the mechanisms of oncogene activation after treatment with hypomethylating agents.

Zinc Finger Protein SALL4 Functions through an AT-Rich Motif to Regulate Gene Expression

Nikki R. Kong et al.

Summary: SALL4 is a zinc finger transcription factor highly expressed in embryonic stem cells and reactivated in aggressive cancers. By identifying its DNA binding domain and consensus binding sequence through experiments, along with discovering new target genes regulated by SALL4 in aggressive liver cancer cells, potential pathways and molecules to target in SALL4-dependent tumors are revealed.

Pseudogene-mediated DNA demethylation leads to oncogene activation

Junsu Kwon et al.

Summary: This study has identified a previously unrecognized role for pseudogenes as potent epigenetic regulators in hepatocellular carcinoma, where they can demethylate and activate oncogenes by interacting with DNMT1. This finding expands our understanding of pseudogenes beyond being considered nonfunctional evolutionary relics.

Summary: This study reports two DICER1-related malignancies with teratoma-like features, expanding their morphologic spectrum and highlighting the multipotentiality of their presumed cell of origin. Furthermore, it describes the first extracranial ETMR with confirmed DICER1 biallelic inactivation and suggests the potential role of other molecular alterations such as KRAS and CTNNB1 mutations in defining the phenotype of embryonal and primitive DICER1-associated neoplasms.
