HuMSC-EV induce monocyte/macrophage mobilization to orchestrate neovascularization in wound healing process following radiation injury

Summary: This study investigates the mechanisms by which human mesenchymal stem cell-derived extracellular vesicles (HuMSC-EV) promote angiogenesis and neovascularization after radiation injury. The study found that HuMSC-EV improved neovascularization of irradiated tissue by stimulating angiogenesis, increasing capillary density, and producing proangiogenic factors. HuMSC-EV also promoted vasculogenesis by recruiting and differentiating bone marrow progenitors. Furthermore, HuMSC-EV enhanced arteriogenesis by mobilizing monocytes and promoting their recruitment to the injured site. These findings suggest that HuMSC-EV could be a potential alternative to stem cells for therapeutic neovascularization in tissue repair.

Mesenchymal Stem Cell Exosomes Promote Functional Osteochondral Repair in a Clinically Relevant Porcine Model

Mechanism for the attenuation of neutrophil and complement hyperactivity by MSC exosomes

Summary: This study identified a feed-forward loop between complements and neutrophils that contributes to the cytokine storm in severe SARS-CoV-2-infected patients. It was observed that the terminal complement activation complex C5b-9 directly triggers neutrophil extracellular trap (NET) release and interleukin (IL)-17 production by neutrophils. Mesenchymal stem cell (MSC) exosomes were found to inhibit this loop by suppressing complement activation through exosomal CD59, suggesting their potential role in alleviating immune dysregulation and inflammation in acute respiratory failure.
