Gelsevirine improves age-related and surgically induced osteoarthritis in mice by reducing STING availability and local inflammation

Meixia Feng et al.

Summary: This study tested the therapeutic effects of gelsevirine on age-related and surgically induced OA in mice and elucidated the underlying mechanisms. The results showed that gelsevirine treatment reduced inflammation and degeneration in chondrocytes, as well as mitigated articular cartilage destruction in OA mice. Mechanistically, gelsevirine targeted STING for ubiquitination and degradation.

L-Arginine based polyester amide/hyaluronic acid hybrid hydrogel with dual anti-inflammation and antioxidant functions for accelerated wound healing

Tong Liu et al.

Summary: This study focuses on the synthesis of arginine-based poly(ester amide) (Arg-PEA) and hyaluronic acid (HA-MA) and their combination into new hybrid hydrogels. The results demonstrate that the hybrid hydrogels show improved internal structure and physicochemical properties with the increase of Arg-PEA content. The study also reveals that the hybrid hydrogels possess anti-inflammatory and antioxidant properties, making them potential scaffold materials for skin regeneration.

Impact of STING Inflammatory Signaling during Intracellular Bacterial Infections

Erika S. Guimaraes et al.

Summary: STING, as an immune sensor, plays a crucial role in the early detection of bacterial pathogens. It activates immune responses by detecting bacterial DNA or cyclic dinucleotides, leading to the production of interferons and proinflammatory cytokines and participating in the metabolic reprogramming of macrophages. However, STING-mediated immune responses during bacterial infections can also be detrimental to the host.

SS-31 ameliorates hepatic injury in rats subjected to severe burns plus delayed resuscitation via inhibiting the mtDNA/STING pathway in Kupffer cells

Yin Wu et al.

Summary: Hepatic injury is common in patients with severe burns plus delayed resuscitation (B + DR), where oxidative stress leads to mitochondrial DNA release and Kupffer cells polarization. SS-31 is an effective antioxidant that prevents hepatic injury by neutralizing reactive oxygen species, inhibiting mtDNA release, protecting hepatocyte mitochondria, suppressing mtDNA/STING pathway activation, and inhibiting pro-inflammatory polarization of Kupffer cells.
