Short-term exposure of 17 alpha-ethynylestradiol impairs water homeostasis by inhibiting osmoregulatory and cortisol regulating genes in adult zebrafish

Summary: The study aimed to investigate the effect of 17 alpha-ethynylestradiol (EE2) on fish osmoregulation. The results showed that EE2 exposure led to symptoms such as body edema, trichangiectasis in the skin, and protrusion of the proptosis in zebrafish. Furthermore, it significantly affected the condition factor, water weight, water content, RBC count, and HCT. Histopathological damages in the gill and kidney were also observed. The study suggests that EE2 exposure may disrupt water homeostasis in zebrafish. Down-regulation of crh, crhr, and acth mRNA levels in the brain was found, consistent with decreased plasma cortisol levels. In addition, elevated expression of aqp3a and aqp1a1 mRNA levels in the gill and decreased expression of atp1a1a.1 and activity of Na+, K+-ATPase in the gill were observed. Suppressed expression of atp1a1a.4 and activity of Na+, K+-ATPase in the kidney were also present. These changes might be due to decreased plasma cortisol levels and low expression of avp in the brain.

Persistent impact of amitriptyline on the behavior, brain neurotransmitter, and transcriptional profile of zebrafish (Danio rerio)

Summary: Discontinuation of amitriptyline (AMI) has been shown to cause long-term withdrawal syndromes in mammals. However, this study focused on the persistent effects of short-term AMI exposure on teleosts. The results showed that AMI exposure led to persistent hypoactivity and abnormal schooling behavior in zebrafish. It also affected the levels of neurotransmitters in the brain and dysregulated certain pathways. These findings highlight the potential long-term adverse effects of AMI on teleosts.
