Spatiotemporal Tracing of the Cellular Internalization Process of Rod-Shaped Nanostructures

Yi-Feng Wang et al.

Summary: This study investigated the internalization behaviors of rod-shaped nanostructures with different aspect ratios using atomic force microscopy, transmission electron microscopy, and molecular dynamic simulation. It revealed that gold nanorods are endocytosed in an intermittent rotation manner and the internalization force and time are positively correlated with the aspect ratios. The study also found that internalization speed is negatively correlated with internalization time.

Synergistic Entry of Individual Nanoparticles into Mammalian Cells Driven by Free Energy Decline and Regulated by Their Sizes

Yushuang Wei et al.

Summary: Cell entry is a common prerequisite for nanomaterial applications. Studies have shown that the synergistic cell entry of nanoparticles is driven by free energy decline and depends on the size of the nanoparticles. Understanding this process can help improve the design of nanostructures for intracellular delivery.

Shutter Effects Enhance Protein Diffusion in Dynamic and Rigid Molecular Networks

Xiaobin Dai et al.

Summary: In this study, a model hydrogel system with structural similarity to ECM was designed using DNA building blocks with rigidity and dynamic binding. The system exhibited enhanced protein diffusion compared to synthetic hydrogels. Experimental and simulation investigations showed a unique transition of diffusion coefficients from a power law to an exponential function with respect to molecular size in this rigid and dynamic network. A shutter model based on the rigidity and dynamic bond exchange was proposed to explain the diffusion mechanism, providing a new perspective on the possible mechanism of enhanced diffusion behaviors in ECM.

Cardiolipin Selectively Binds to the Interface of VsSemiSWEET and Regulates Its Dimerization

Zhixiong Deng et al.

Summary: This study used molecular dynamics simulations to uncover the key role of interfacial cardiolipin in regulating the functional dimerization of the membrane protein VsSemi-SWEET. The research identified four binding sites for cardiolipins and observed two types of dimerization modes during the assembly process. The findings provide valuable information on the relationship between lipid binding and functional dimerization, contributing to a better understanding of the molecular mechanism of sugar transporter biological function.

Cholesterols Work as a Molecular Regulator of the Antimicrobial Peptide-Membrane Interactions

Jia Li et al.

Summary: The study demonstrates that existing cholesterols in animal cell membranes can inhibit the membrane poration activity of melittin. This inhibitory effect is attributed to the accumulation of Chol molecules around the membrane-bound peptide chains, hindering further insertion of peptides into the membrane. These findings suggest the potential to develop targeted drugs based on the membrane component specificity between bacterial and animal cells.

Summary: This study investigated the bystander uptake effect of FNPs on BNPs at the cell membrane level, showing that co-administered FNPs can enhance the membrane internalization efficiency of BNPs by enhancing the membrane-curvature-mediated attraction and aggregation of NPs. Suggestions were given to modulate the BNP internalization through controlling the FNP properties such as size, concentration and surface-ligand density.
