Differentiating Catalysis in the Dearomative [4+2]-Cycloaddition Involving Enals and Heteroaromatic Aldehydes

Aleksandra Topolska et al.

Summary: The application of differentiating catalysis in the [4 + 2]-cycloaddition reaction between 2-alkyl-3-formylheteroarenes and alpha,beta-unsaturated aldehydes is described in this paper. The use of the same aminocatalyst allows for independent activation of both starting materials, achieving high enantio- and diastereoselectivity through LUMO-lowering and HOMO-rising principles. The combination of dearomative dienamine activation with iminium ion chemistry further improves the outcome of the process.

Rings in Clinical Trials and Drugs: Present and Future

Jonathan Shearer et al.

Summary: In this study, we conducted a comprehensive analysis of ring compounds and FDA-approved drugs. We found that 67% of small molecules in clinical trials only utilize ring systems that are already present in marketed drugs. Additionally, there are approximately 450,000 unique ring systems derived from synthesized chemical space, but molecules in clinical trials represent only 0.1% of this pool. Furthermore, by making systematic changes to existing drug and clinical trial ring systems, we were able to predict potential future clinical trial ring systems that could cover a significant portion of novel ring systems entering clinical trials.

Aminocatalytic Synthesis of Uracil Derivatives Bearing a Bicyclo[2.2.2]octane Scaffold via a Doubly Cycloadditive Reaction Cascade

Maciej Saktura et al.

Summary: The study presents a method for aminocatalytic synthesis of highly enantiomerically enriched uracil derivatives with a bicyclo[2.2.2]octane scaffold, utilizing various aminocatalytic activation strategies for stereoselective construction. The reaction cascade involves doubly cycloadditive steps with dearomatization of the uracil moiety, and excellent stereoselectivity is achieved using 2-(diphenylmethyl)pyrrolidine as aminocatalyst.

Direct, Asymmetric Synthesis of Carbocycle-Fused Uracils via [4+2] Cycloadditions: a Noncovalent Organocatalysis Approach

Enrico Marcantonio et al.

Summary: The remote enolization strategy, strategic exploitation of noncovalent bifunctional organocatalysis, and efficient stereoselective trapping of dienolate intermediates with nitroolefins led to the successful synthesis of functionalized, chiral carbocycle-fused uracils with three contiguous stereocenters in one step, showing good levels of enantioselectivity and complete diastereocontrol. Furthermore, the enantiopure products could be obtained via simple one-cycle recrystallizations and the scaffolds could be further functionalized without losing their chiral integrity.
