Synthesis of Stereoisomeric Simplified Analogs of Alotaketals toward the Elucidation of the Structural Requirements of Protein Kinase C Isozyme-Selective Binding

Summary: A set of four stereoisomeric compounds were designed and synthesized as ligands of protein kinase C (PKC). The compounds were simplified analogs of the alotaketals, a class of natural products that were predicted to be ligands of PKC by computational screening. Bioassays revealed that the orientation of the alkyl side chain of the analogs was important for PKC binding and that the stereochemistry of the fused ring moiety influenced the PKC isozyme selectivity.

Isolation, Synthesis, and Structure-Activity Relationship Study on Daphnane and Tigliane Diterpenes as HIV Latency-Reversing Agents

Summary: Three new diterpenes were isolated from the roots of Stellera chamaejasme L., and their structures were determined through NMR and mass spectroscopic analyses. Compounds 1 and 2 are the first derivatives containing a hydroxy group at C-2 in the family of daphnane and tigliane diterpenes. Compound 1, with a chlorine atom, and compound 3, with an odd-number acyl group, are unique plant metabolites.

Ansellone J, a Potent in Vitro and ex Vivo HIV-1 Latency Reversal Agent Isolated from a Phorbas sp. Marine Sponge

Summary: Five new minor sesterterpenoids have been isolated from a marine sponge collected in British Columbia, and their structures have been determined. Two of these compounds, ansellone J (6) and phorone C (8), exhibit potent activity in reversing HIV-1 latency in vitro, with ansellone J (6) showing similar potency to prostratin (3) at a 10-fold lower concentration.

Summary: The total synthesis and biological evaluation of the marine sesterterpenoid ansellone A, along with its analogues, were successfully carried out. The key to the success of the synthetic route was the Prins cyclization reaction enabled by the strategic use of the TfO group to stabilize the acid-labile tertiary allylic alcohol. The structure-activity relationship study indicated that the alcohol analogue exhibited more potent activity than compound 1.
