Continuous-Flow Photocatalysis for the Direct C-H Trifluoromethylation of Heterocycles with an Organic Photoredox Catalyst

Jianwen Yang et al.

Summary: Continuous-flow photocatalytic direct C-H trifluoromethylation of heterocycles was achieved using an organic photoredox catalyst: 2,4,6-tris(diphenylamino)-3,5-difluorobenzonitrile (3DPA2FBN). By combining organic photoredox catalysis and continuous-flow techniques, various trifluoromethylated heterocycles were rapidly synthesized with up to 85% yield and 80 min residence time under metal- and oxidant-free reaction conditions.

Electron-Deficient Fluoroarene-Mediated Synthesis of Trifluoromethyl Ketones from Carboxylic Acids

Madhukar S. Said et al.

Summary: This study reveals a method for the synthesis of trifluoromethyl ketones via the fluoroarene-mediated trifluoromethylation of carboxylic acids. The fluoroarene activates the acid group and generates the fluoride source in situ for the trifluoromethylation reaction. The protocol is safe, metal-free, operates under mild conditions, and does not require external additives to generate trifluoromethyl anion. The transformation shows good functional group tolerance and delivers high yields of trifluoromethyl ketones in both batch and continuous flow processes.

KHMDS/Triglyme Cryptate as an Alternative to Phosphazene Base in Stereodivergent Pentafluoroethylation of N-Sulfinylimines Using HFC-125

Yuji Sumii et al.

Summary: A protocol for the stereodivergent pentafluoroethylation of N-sulfinylimines has been developed using HFC-125 with KHMDS/triglyme. The presence or absence of triglyme enables the selective synthesis of both diastereomers of the pentafluoroethylated amines. This additive-controlled protocol offers a straightforward and cost-effective alternative to the previously reported base-controlled stereodivergent trifluoromethylation using potassium hexamethyldisilazide (KHMDS) versus P4-tBu.

Pentafluoroethylation of Carbonyl Compounds by HFC-125 via the Encapsulation of the K Cation with Glymes

Yamato Fujihira et al.

Summary: A simple protocol using potassium bases with triglyme or tetraglyme as solvents can safely yield pentafluoroethylation products. Encapsulation of the K cation by glymes inhibits its contact with reactive anionic pentafluoroethyl counterions, preventing their transformation into explosive tetrafluoroethylene. The generation of sterically demanding K(G3)(2) and K(G4)(2) cations serves as an effective reservoir for unstable pentafluoroethyl anions.
