Novel primers for 16S rRNA gene-based archaeal and bacterial community analysis in oceanic trench sediments

Summary: This study designed specific primers to improve the detection resolution of unique microbial groups in oceanic trench sediments. Through high-throughput sequencing analysis, the V3-V4 hypervariable region was found to be the most informative and representative for oceanic trench microbial groups. Compared to universal primers, the newly designed primers were able to detect more microbial families, especially archaeal ones, resulting in the discovery of more species that were previously ignored in universal primer-based experiments.

Fast development of microbial cultures for the anaerobic oxidation of CH4 coupled to denitrification employing widely available inocula

Summary: This study assessed two widely available inocula for the anaerobic oxidation of methane coupled with NO3 reduction. It found that methane consumption via this process could be observed within a relatively short period using secondary activated sludge and a mixture of activated sludge and anaerobic granular sludge as inoculum. The study also identified the dominant microbial communities in the cultures and confirmed the absence of nitrous oxide, an undesirable intermediate, at the end of the incubations. The findings suggest that N-AOM processes can be implemented quickly and easily, making it a viable option for the treatment of residual methane in wastewater treatment plants and waste management facilities.

Impact of an anoxic desulfurization process on methane content of the purified biogas

The reduction in CO2 concentration and increase in CH4 concentration in the purified biogas were found to be proportional, with CO2 absorption playing a role in this process. Additionally, the presence of methane-oxidizing bacteria suggests that methane oxidation occurred in the desulfurization system.

Denitrifying anaerobic methane oxidation (DAMO) cultures: Factors affecting their enrichment, performance and integration with anammox bacteria

Summary: The recently discovered process of denitrifying anaerobic methane oxidation (DAMO) connects the carbon and nitrogen biogeochemical cycles by linking the anaerobic oxidation of methane to denitrification. The DAMO process shows potential for reducing the greenhouse effect through the assimilation of dissolved methane, and offers a new perspective on denitrification through methane oxidation rather than organic matter. The two main microorganisms responsible for DAMO, M. oxyfera and M. nitroreducens, exist together in nature through a syntrophic relationship and may coexist with anaerobic ammonium oxidation (Anammox) bacteria, presenting opportunities for improved nitrogen removal systems and wastewater treatment alternatives.
