Mitochondrial Dysfunction: A Prelude to Neuropathogenesis of SARS-CoV-2

Artem Pliss et al.

Summary: The SARS-CoV-2 virus can cause neurological conditions by invading the nervous system. The dysfunction of mitochondria in brain microglia cells is believed to play a role in the neuropathology of SARS-CoV-2. Using Raman microspectrometry, researchers found that infected microglia cells exhibited reduced mtDNA content and increased phospholipid saturation levels in mitochondria. These changes may be linked to increased synthesis of reactive oxygen species in the organelles.

Interaction of Mitochondrial Calcium and ROS in Neurodegeneration

Artyom Y. Baev et al.

Summary: Neurodegenerative disorders are devastating diseases characterized by the loss of neurons in specific brain regions. Recent research has identified genes, toxins, and risk factors associated with these diseases, and has found similarities in the involvement of mitochondria, oxidative stress, and abnormal calcium signaling in neurons and astrocytes.

Lipidome Alterations following Mild Traumatic Brain Injury in the Rat

Eric C. Gier et al.

Summary: This research identified lipid biomarker candidates for traumatic brain injury (TBI) by analyzing serum samples from Sprague Dawley rats. Machine learning and feature selection approaches were used to construct lipid panels capable of distinguishing injured and uninjured rats with over 90% accuracy. These biomarker candidates were associated with various metabolic pathways.

Blast-induced injury responsive relative gene expression of traumatic brain injury biomarkers in human brain microvascular endothelial cells

Rebecca R. Schmitt et al.

Summary: Disruption of the blood-brain barrier is crucial in the progression of traumatic brain injury, and further research into the mechanism is necessary for understanding TBI pathophysiology. Studying gene expression changes in HBMVEC can help elucidate the interaction between BBB pathophysiology and TBI.

IDH1 mutations induce organelle defects via dysregulated phospholipids

Adrian Lita et al.

Summary: This study reveals that IDH1(mut) results in an increase in monounsaturated fatty acids and their phospholipids in the endoplasmic reticulum, leading to Golgi and ER dilation. Inhibition of IDH1 mutation or SCD silencing restores ER and Golgi morphology, while D-2HG and oleic acid induces morphological defects in these organelles. Moreover, the overexpression of SCD induced by IDH1(mut) rearranges the distribution of lipids in the organelles of glioma cells, contributing to potential therapeutic implications.

Repeated mild traumatic brain injuries perturb the mitochondrial biogenesis via DNA methylation in the hippocampus of rat

Nagalakshmi Balasubramanian et al.

Summary: The study found that repeated mild traumatic brain injury (rMTBI) results in hypermethylation at the TFAM promoter and hypomethylation at mitochondrial DNA (mtDNA) promoters (HSP1 and HSP2), leading to impaired mitochondrial biogenesis and reduced ATP levels in the hippocampus. Treatment with methionine, a methyl donor, restored methylation levels on mtDNA promoters and normalized mitochondrial gene expression and ATP activity.

Interleukin-1β mediates alterations in mitochondrial fusion/fission proteins and memory impairment induced by amyloid-β oligomers

Andre F. Batista et al.

Summary: The study found that IL-1 beta mediates the impact of A beta Os on proteins involved in mitochondrial dynamics, and suggests that strategies aimed at preventing pathological alterations in these proteins may counteract synapse loss and cognitive impairment in Alzheimer's disease (AD).

A Single-Organelle Optical Omics Platform for Cell Science and Biomarker Discovery

Artem Pliss et al.

Summary: The Ramanomics platform enables molecular profiling of single organelles in live-cell environments using confocal Raman microspectrometry and biomolecular component analysis algorithms. Significant variations in organellar composition between different cell lines have been discovered, and Ramanomics has been proven effective in identifying diseased cells and revealing large-scale molecular transformations accompanying disease development in organelles.

Oxidative Stress Signaling in Blast TBI-Induced Tau Phosphorylation

Chunyu Wang et al.

Summary: The study showed that the initial increase in tau phosphorylation following a single mild blast was associated with oxidative damage, with the activation of ERK and JNK likely mediating the effects of oxidative stress on tau phosphorylation.

Postoperative Rise of Circulating Mitochondrial DNA Is Associated with Inflammatory Response in Patients following Pancreaticoduodenectomy

Niv Pencovich et al.

Summary: Following pancreaticoduodenectomy, some patients showed a significant increase in circulating mtDNA levels, which were associated with postoperative fever, decreased levels of hemoglobin and albumin, increased white blood cell counts, and delayed gastric emptying. These patients also had a poorer postoperative course.
