Optimizing memory performance and emotional states: multi-level enhancement of adult hippocampal neurogenesis

Ya-Dong Li et al.

Summary: Adult hippocampal neurogenesis (AHN) is crucial for memory and emotion processing. In this review, we summarize recent studies on how different aspects of AHN, such as the number, maturation state, and activity of adult-born neurons (ABNs), contribute to hippocampal function. We propose a multi-level enhancement approach that combines increased number, elevated activity, and enhanced maturation of ABNs to optimize overall hippocampal performance. Additionally, the integration of ABNs leads to significant remodeling of local hippocampal circuits and modulates brain-wide network dynamics during behavior.

Altered adult neurogenesis and gliogenesis in patients with mesial temporal lobe epilepsy

Aswathy Ammothumkandy et al.

Summary: In individuals with epilepsy, the longer the duration of the disease, the fewer new neurons are produced and the greater the presence of immature astrocytes. Immature neurons are mostly inactive in cases with epilepsy, while immature astrocytes persist and their activity is associated with neuronal hyperactivity.

Hypothalamic modulation of adult hippocampal neurogenesis in mice confers activity-dependent regulation of memory and anxiety-like behavior

Ya-Dong Li et al.

Summary: Adult hippocampal neurogenesis is critical for memory and emotion processing. Researchers have found that chronic patterned optogenetic stimulation of SuM neurons in the hypothalamus promotes neurogenesis and modulates behavior. SuM neurons are highly responsive to environmental novelty.

Molecular landscapes of human hippocampal immature neurons across lifespan

Yi Zhou et al.

Summary: This study identified molecular characteristics of immature dentate granule cells (imGCs) in the adult human hippocampus using single-nucleus RNA sequencing, revealing common molecular hallmarks across the lifespan and age-dependent transcriptional dynamics. The study also showed a decreased number of imGCs in Alzheimer's disease and demonstrated neurogenesis in the adult human hippocampus.

Transcriptome dynamics of hippocampal neurogenesis in macaques across the lifespan and aged humans

Wei Wang et al.

Summary: This study investigates the cellular heterogeneity and molecular characteristics of the hippocampi in macaques and aged humans using droplet-based single-nucleus RNA sequencing (snRNA-seq). The findings reveal the dynamics of the neurogenic lineage and the diversity of astrocytes and microglia in the hippocampus. Primate-specific markers are identified and their functions are validated. Additionally, active astrocytes and microglia with proinflammatory responses are observed in aged samples, suggesting their contribution to the decrease and variability of adult hippocampal neurogenesis.

Ventral pallidal GABAergic neurons control wakefulness associated with motivation through the ventral tegmental pathway

Ya-Dong Li et al.

Summary: VP GABAergic neurons play a crucial role in regulating arousal by modulating dopaminergic neurons and promoting wakefulness. Additionally, they promote arousal through innervation of the lateral hypothalamus, but do not affect depression-like behaviors or food intake.

Restoring miR-132 expression rescues adult hippocampal neurogenesis and memory deficits in Alzheimer's disease

Hannah Walgrave et al.

Summary: Neural stem cells in the hippocampal neurogenic niche play a crucial role in lifelong neurogenesis, while miR-132 has been identified as a significant regulator affecting hippocampal neurogenesis in Alzheimer's disease. Research demonstrates that replacing miR-132 can restore neurogenesis and memory deficits in the hippocampus of AD mice, highlighting the potential therapeutic efficacy of targeting miR-132 in neurodegenerative diseases.

Impact of neurodegenerative diseases on human adult hippocampal neurogenesis

J. Terreros-Roncal et al.

Summary: Disrupted hippocampal performance is a major factor contributing to psychiatric comorbidities and cognitive impairments in patients with neurodegenerative disorders. Adult hippocampal neurogenesis (AHN) plays a crucial role in several neurodegenerative diseases, with abnormalities found in adult-born dentate granule cells. Aging and diseases alter the neurogenic niche, reduce microglial phagocytic capacity, trigger astrogliosis, and change the microvasculature of the dentate gyrus, leading to enhanced vulnerability of AHN to neurodegeneration.

Supramammillary regulation of locomotion and hippocampal activity

Jordan S. Farrell et al.

Summary: Research has identified neurons in the supramammillary nucleus (SuM) that are highly correlated with future locomotor speed and reliably drive locomotion. Specifically, Tac1-expressing neurons in the SuM were found to control the activity of speed-modulated hippocampal neurons. These findings suggest that the SuM not only regulates basic locomotor activity but also selectively shapes hippocampal neural activity in a way that may support spatial navigation.

Modulating adult neurogenesis affects synaptic plasticity and cognitive functions in mouse models of Alzheimer's disease

Xiaoqin Zhang et al.

Summary: Research indicates that removing adult neural stem cells can improve memory in AD mouse models, while exposure to enriched environments can promote adult neurogenesis and ameliorate memory deficits in APP/PS1 mice. Both inhibiting abnormal adult neurogenesis and enhancing healthy adult neurogenesis could be beneficial for AD, and are not mutually exclusive.

Microglia use TAM receptors to detect and engulf amyloid β plaques

Youtong Huang et al.

Summary: The TAM receptor kinases are found to be essential for microglial recognition and engulfment of amyloid plaques in Alzheimer's disease, with their phagocytosis actually promoting the formation of dense-core plaques. This study also reveals that Axl and Mer play crucial roles in microglial response to amyloid plaques, and their deficiency leads to impaired detection, response, organization, and phagocytosis of amyloid-beta plaques.
