Neuronal activity induces glucosylceramide that is secreted via exosomes for lysosomal degradation in glia

Summary: Recessive variants in GBA1 can lead to Gaucher disease, a common lysosome storage disease. Using CRISPR-Cas9, we generated a null allele of the Drosophila ortholog Gba1b and found that it is expressed in glia but not in neurons. GlcCer is synthesized upon neuronal activity and transported from neurons to glia through exosomes. Glial TGF-beta/BMP induces GlcCer transfer from neurons to glia, and the White protein promotes GlcCer trafficking to glial lysosomes for degradation.

Regional heterogeneity of astrocyte morphogenesis dictated by the formin protein, Daam2, modifies circuit function

Summary: Loss of the formin protein Daam2 in astrocytes leads to increased morphological complexity in the cortex and olfactory bulb, but has opposing effects on astrocytic calcium dynamics. These differential physiological effects result in increased excitatory synaptic activity in the cortex and increased inhibitory synaptic activity in the olfactory bulb, altering olfactory behaviors. Proteomic profiling and immunoprecipitation experiments reveal Slc4a4 as a binding partner of Daam2 in the cortex, and deletion of both Daam2 and Slc4a4 restores the morphological alterations seen in Daam2 mutants.

ABCD1 and X-linked adrenoleukodystrophy: A disease with a markedly variable phenotype showing conserved neurobiology in animal models

Summary: X-ALD is a phenotypically heterogeneous disorder caused by a mutation in the ABCD1 gene, leading to defective peroxisomal beta-oxidation of VLCFAs. The disease presents with a variety of clinical manifestations and lacks genotype-phenotype correlation. Elevated VLCFAs in blood serve as a biochemical marker for diagnosis, while investigational gene therapy approaches show promise for improved outcomes. However, the pathophysiological mechanisms of the disease remain poorly understood, limiting targeted therapeutic options. Animal models recapitulate some aspects of the disease, but the lack of cerebral invasion of leukocytes and demyelination in these models highlights the need for further research to uncover still unknown pathological processes.

Neurotoxic reactive astrocytes induce cell death via saturated lipids
