Treatment landscape of triple-negative breast cancer - expanded options, evolving needs

Giampaolo Bianchini et al.

Summary: Recent advances in omics technologies have improved our understanding of the heterogeneity and microenvironment of triple-negative breast cancers (TNBCs), supporting a view of this breast cancer subtype as an ecosystem that encompasses both intrinsic and extrinsic features of cancer cells. The authors of this Review describe the current and upcoming therapeutic landscape of TNBC and discuss how an integrated view of the TNBC ecosystem can provide improved opportunities for tailoring treatment.

FOXM1 Inhibition Enhances the Therapeutic Outcome of Lung Cancer Immunotherapy by Modulating PD-L1 Expression and Cell Proliferation

Hamadi Madhi et al.

Summary: The focus of this study is FOXM1, which is identified as a potential therapeutic target for cancer immunotherapy and associated with the modulation of PD-L1 expression. The study found that selective knockdown of FOXM1 or treatment with TST significantly reduces PD-L1 expression in NSCLC cells and inhibits proliferation. Animal studies showed that TST treatment downregulates PD-L1 expression in NSCLC tumors and reduces tumor size without side effects. Combined treatment with TST and anti-4-1BB antibody induces synergistic therapeutic outcomes against immune resistant lung tumors and increases the number of CD3(+) T cells in tumor tissues.

USP7 Induces Chemoresistance in Triple-Negative Breast Cancer via Deubiquitination and Stabilization of ABCB1

Yueh-Te Lin et al.

Summary: This study identified USP7 as a potential therapeutic target for overcoming chemoresistance in triple-negative breast cancer. The research demonstrated that USP7 promotes chemoresistance in TNBC by stabilizing the ABCB1 protein, providing insights into targeted therapy for TNBC patients with acquired resistance.
