Cancer statistics, 2022

Rebecca L. Siegel et al.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Multicentre, prospective study on local treatment of metastatic prostate cancer (LoMP study)

Sarah Buelens et al.

Summary: The study aimed to investigate the role of cytoreductive radical prostatectomy in patients with newly diagnosed metastatic prostate cancer. The results showed longer castration resistant cancer-free survival and higher local event-free survival in the surgery group. However, there was no significant difference between treatment groups in the multivariable analysis.

The promising role of new molecular biomarkers in prostate cancer: from coding and non-coding genes to artificial intelligence approaches

Ana Paula Alarcon-Zendejas et al.

Summary: This review provides an overview of recent advances in the discovery of new molecular biomarkers through transcriptomics, genomics, and artificial intelligence for the improvement of clinical management in prostate cancer. The identification of novel biomarkers is expected to contribute to earlier and more accurate diagnosis, ultimately improving patient prognosis and quality of life. Bioinformatic approaches such as machine learning and deep learning also offer potential for clinical decision-making and personalized management in prostate cancer.

Benchmarking weakly-supervised deep learning pipelines for whole slide classification in computational pathology

Narmin Ghaffari Laleh et al.

Summary: This study systematically compared the performance differences between classical weakly-supervised analysis methods and multiple-instance learning methods in clinically relevant prediction tasks. The results showed that all methods performed well in the histological tumor subtyping of renal cell carcinoma, while classical weakly-supervised methods outperformed MIL-based methods in mutation prediction tasks for colorectal, gastric, and bladder cancer.

Atlas of clinically distinct cell states and ecosystems across human solid tumors

Bogdan A. Luca et al.

Summary: This study introduced EcoTyper, a machine learning framework for identifying cell states and multicellular communities from gene expression data, and identified 69 transcriptionally defined cell states in 12 major cell lineages across 16 types of human carcinoma. By analyzing cell-state co-occurrence patterns, it discovered ten clinically distinct multicellular communities, including those linked to adverse survival.

Next Generation Imaging Techniques to Define Immune Topographies in Solid Tumors

Violena Pietrobon et al.

Summary: Cancer immunotherapy shows great promise in hematological malignancies, but its efficacy in solid tumors is limited due to immune exclusion mediated by various barriers. There is currently no unified understanding of the role of different models of immune exclusion in solid tumors. Mapping immune landscapes in cancers of different types is crucial to understand the distribution of lymphocytes in the tumor microenvironment.

Data-efficient and weakly supervised computational pathology on whole-slide images

Ming Y. Lu et al.

Summary: The CLAM method utilizes attention-based learning to identify subregions with high diagnostic value for accurate classification of whole-slide images. It can localize well-known morphological features without the need for spatial labels, outperforming standard weakly supervised classification algorithms, and adapt to independent test cohorts, smartphone microscopy, and varying tissue content.

Network-Based Analysis to Identify Drivers of Metastatic Prostate Cancer Using GoNetic

Louise de Schaetzen van Brienen et al.

Summary: To identify rare driver genes in metastatic prostate cancer, researchers introduced a novel network-based method called GoNetic which successfully prioritized known drivers and rare driver candidates. This method utilizes sample-specific mutations and the prior interaction network, showing promise in identifying potential driver genes for disease progression.

AI-based pathology predicts origins for cancers of unknown primary

Ming Y. Lu et al.

Summary: Cancer of unknown primary (CUP) is a difficult diagnosis as the primary site of tumor origin cannot be determined. The deep-learning-based algorithm TOAD provides a differential diagnosis for the origin of the primary tumor, achieving high accuracy on test sets and reducing the occurrence of CUP by assisting in assigning differential diagnoses for complicated cases.

A Subset of Localized Prostate Cancer Displays an Immunogenic Phenotype Associated with Losses of Key Tumor Suppressor Genes

Carla Calagua et al.

Summary: A subset of primary prostate cancer expresses PD-L1 and exhibits immunogenic characteristics. Immunogenic prostate cancer shows comparable CD8(+) T-cell content with renal cell carcinoma, potentially driven by genomic losses of RBI, BRCA2, and CHD1.
