Insights into the Steps of Breast Cancer-Brain Metastases Development: Tumor Cell Interactions with the Blood-Brain Barrier

Summary: Brain metastases (BM) is an increasing problem for breast cancer (BC) patients, and recent studies have shown that the BC molecular subtype has a strong impact on BM development. This study investigates the interaction between BC cells of different molecular subtypes and the blood-brain barrier (BBB). The results show that TNBC cells have a greater ability to influence the BBB in vitro and develop BM in vivo compared to luminal BC cells. Transcriptome analysis reveals potential key regulators involved in the processes of angiogenesis and cell migration and invasion. The study suggests that the ability of tumor cells to disrupt the BBB through angiogenesis activation and increased cellular motility is necessary for BC cells to overcome the BBB and develop brain metastases.

Interplay between desmoglein2 and hypoxia controls metastasis in breast cancer

Summary: High expression of DSG2 promotes tumor growth, increases the prevalence of CTC clusters, and facilitates distant organ colonization. The dynamic regulation of DSG2 by hypoxia, with down-regulation in hypoxic regions of primary tumors leading to elevated EMT gene expression, allows cells to detach from the primary tumor and undergo intravasation. The derepression of DSG2 after intravasation and release of hypoxic stress is associated with increased ability to colonize distant organs, mediated by Hypoxia-Induced Factor1 alpha (HIF1 alpha).

Detection of clustered circulating tumour cells in early breast cancer
