Analysis of human leukocyte antigen associations in human papillomavirus-positive and -negative head and neck cancer: Comparison with cervical cancer

Chameera Ekanayake Weeramange et al.

Summary: The risk of human papillomavirus (HPV)-positive head and neck cancer (HNC) is associated with distinct human leukocyte antigen (HLA) variants, some of which are shared by both cervical cancer and HPV-positive HNC. Several previous genome-wide association studies were replicated in this study, but the associations were limited to the HPV-positive HNC group.

Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Organoid Models for Precision Cancer Immunotherapy

Cai-Ping Sun et al.

Summary: Cancer immunotherapy modulates the immune system to treat diseases, but conventional animal and in vitro models fail to accurately simulate the tumor immune microenvironment. More physiomimetic cancer models, such as patient-derived organoids, are needed to evaluate the efficacy of immunotherapy agents. The dynamic interactions between neoplastic cells and non-neoplastic host components in the tumor immune microenvironment play a crucial role in carcinogenesis, tumor metastasis, cancer progression, and drug resistance. Tumor organoids can effectively recapitulate the tumor immune microenvironment and be used for testing immunotherapy agents and personalized cancer immunotherapy.

Cisplatin for cancer therapy and overcoming chemoresistance

Ranmali Ranasinghe et al.

Summary: Cisplatin is a leading chemotherapy drug for cancer treatment, but its acute toxicity remains a major challenge. While efforts are ongoing to discover more effective drugs, nothing has surpassed the benefits of cisplatin so far. Oxaliplatin is considered superior in colorectal cancer, but cisplatin still has a wide application in treating gynaecological cancers.

Roles of mesenchymal stromal cells in the head and neck cancer microenvironment

Marcelo Coutinho de Miranda et al.

Summary: Head and neck cancer (HNC) is a common malignancy with high morbidity and mortality rates worldwide. The tumor microenvironment, which includes various cells regulating tumor development, plays a role in cancer progression and treatment response. Understanding the role of individual cells in tumor progression, particularly mesenchymal stromal cells (MSCs), remains a challenge due to the lack of specific markers and similarities with other stromal cells.
