Latency-associated Peptide Identifies Immunoevasive Subtype Gastric Cancer With Poor Prognosis and Inferior Chemotherapeutic Responsiveness

Yifan Cao et al.

Summary: This study revealed the clinical significance of LAP in predicting survival outcomes and chemotherapeutic responsiveness in gastric cancer. High LAP expression was associated with poor overall survival and inferior response to adjuvant chemotherapy. LAP was also associated with an immunoevasive tumor microenvironment and dysfunctional CD8(+) T cell infiltration.

The evolving role of immune cells in prostate cancer

Chao Wang et al.

Summary: Prostate cancer is a common cancer among men in Western countries, and the standard therapy of ADT may lead to resistance and tumor progression. Recent data suggests that immune cells play important roles in the initiation, progression, metastasis, and treatment of prostate cancer.

Androgen receptor activity in T cells limits checkpoint blockade efficacy

Xiangnan Guan et al.

Summary: Immune checkpoint blockade has revolutionized oncology by inducing durable anti-tumour immunity. In advanced prostate cancer patients, immunotherapy treatments have largely failed. However, inhibiting androgen receptor activity can increase the sensitivity of tumor-bearing hosts to checkpoint blockade therapy by enhancing CD8 T cell function and improving responsiveness to PD-1 targeted therapy.

Androgen conspires with the CD8+ T cell exhaustion program and contributes to sex bias in cancer

Hyunwoo Kwon et al.

Summary: This study reveals a sex bias in the development and progression of nonreproductive organ cancers, with male patients having a higher frequency of CD8(+) T cells lacking effector activity. This bias is driven by gonadal androgen and not by sex chromosomes. The androgen receptor plays a crucial role in promoting CD8(+) T cell exhaustion through a sex-specific regulon and acts as a direct transcriptional transactivator of Tcf7/TCF1. Inhibiting the androgen receptor rewires the tumor microenvironment, favoring effector T cell differentiation and enhancing the efficacy of anti-PD-1 immune checkpoint blockade.

Androgen receptor-mediated CD8+ T cell stemness programs drive sex differences in antitumor immunity

Chao Yang et al.

Summary: This study reveals the immunological mechanism underlying sexual differences in non-reproductive cancers, showing that male CD8(+) T cells have impaired effector and stem cell-like properties compared to female CD8(+) T cells. Androgen receptor inhibits the activity and sternness of male tumor-infiltrating CD8(+) T cells, while castration combined with anti-PD-L1 treatment restricts tumor growth in male mice. In humans, male CD8(+) T cells have fewer stem cell-like memory cells, and AR expression correlates with CD8(+) T cell exhaustion in cancer patients. These findings provide insights into sex-based immunotherapeutic strategies for cancer treatment.

CDK12 Deficiency and the Immune Microenvironment in Prostate Cancer

Tamara L. Lotan et al.

Summary: CDK12 inactivation in prostate cancer may generate fusion-associated neoantigens and elicit immune responses. The T-cell immune microenvironment of CDK12-deficient prostate cancers showed an increase in immunosuppressive CD4(+) FOXP3(-) T cells compared to CDK12-proficient controls.

Single-cell analysis reveals transcriptomic remodellings in distinct cell types that contribute to human prostate cancer progression

Sujun Chen et al.

Summary: Chen et al. conducted single-cell analysis to identify transcriptomic remodeling in various cell types that regulate prostate cancer progression. They found that the tumor microenvironment displayed activation of progression-associated transcriptomic programs and identified a subset of endothelial cells that actively communicate with tumor cells, promoting cancer cell invasion. Their study also revealed promiscuous KLK3 expression and the ability of cancer cells to alter T-cell transcriptomes.
