Cardiac Safety of Kinase Inhibitors - Improving Understanding and Prediction of Liabilities in Drug Discovery Using Human Stem Cell-Derived Models

Ricarda Ziegler et al.

Summary: This study focused on integrating and measuring ATP, apoptosis dynamics, and functional cardiac indexes in human stem cell-derived cardiomyocytes to gain insights into the molecular determinants of compromised cardiac function caused by small molecule kinase inhibitors (SMKIs). The results showed that some SMKIs significantly affected cardiac function, especially those involved in the PI3K-AKT pathway. Pearson's correlation analysis indicated a good correlation between different functional read-outs, suggesting that measuring ATP concentrations and apoptosis in vitro could provide important insights into the mechanisms of cardiotoxicity.

Long-term outcomes with frontline nilotinib versus imatinib in newly diagnosed chronic myeloid leukemia in chronic phase: ENESTnd 10-year analysis

Hagop M. Kantarjian et al.

Summary: In patients with newly diagnosed chronic myeloid leukemia, nilotinib demonstrated higher cumulative molecular response rates, lower rates of disease progression and CML-related death, and increased eligibility for treatment-free remission compared to imatinib. However, nilotinib was associated with higher cumulative rates of cardiovascular events, highlighting the importance of carefully assessing the benefit-risk profile when considering its use as frontline therapy.

Etiology-Dependent Impairment of Diastolic Cardiomyocyte Calcium Homeostasis in Heart Failure With Preserved Ejection Fraction

Summary: HFpEF patients showed increased t-tubule density compared to controls, while HFrEF patients had reduced t-tubule density. This suggests distinct cardiomyocyte phenotypes underlie these heart failure entities.

Endoplasmic reticulum stress promotes sorafenib resistance via miR-188-5p/hnRNPA2B1-mediated upregulation of PKM2 in hepatocellular carcinoma

Bei Zhou et al.

Summary: The study revealed that endoplasmic reticulum stress promotes sorafenib resistance in hepatocellular carcinoma via the upregulation of PKM2 through the miR-188-5p/hnRNPA2B1 pathway. Targeting this pathway and ER stress could be a potential strategy to overcome sorafenib resistance in HCC treatment.
