LSD1-mediated stabilization of SEPT6 protein activates the TGF-β1 pathway and regulates non-small-cell lung cancer metastasis

Yanni Hong et al.

Summary: The study demonstrates that LSD1 promotes proliferation, migration, and invasion of NSCLC cells by demethylating the SEPT6 promoter and suppressing apoptosis. Additionally, LSD1-mediated SEPT6 accelerates NSCLC metastasis in vivo through the TGF-beta 1/Smad pathway.

I-M Launonen et al.

Summary: In this study, the authors used highly multiplexed imaging to analyze the immune microenvironment of high-grade serous ovarian cancers (HGSOC) and identified phenotypic characteristics and spatial interactions associated with BRCA1/2 gene mutations. These findings have important implications for improving immunotherapeutic strategies and patient stratification.

Chromatin state dynamics confers specific therapeutic strategies in enhancer subtypes of colorectal cancer

Elias Orouji et al.

Summary: Enhancer aberrations are key epigenetic features in colorectal cancers, with active enhancer dynamics underlying tumor progression and potential for precision combination therapy. By mapping chromatin profiles and identifying distinct enhancer subtypes, combinatorial therapeutic strategies targeting specific pathways based on enhancer patterns have been proposed for different CRC subgroups, potentially improving patient outcomes.

LSD1 deletion decreases exosomal PD-L1 and restores T-cell response in gastric cancer

Dan-Dan Shen et al.

Summary: The expression of LSD1 is significantly elevated in gastric cancer patients and is associated with PD-L1 expression. LSD1 inhibits the response of T cells in the gastric cancer microenvironment by inducing the accumulation of PD-L1 in exosomes. Exosomes can also serve as vehicles for LSD1 to regulate T cell immunity in other cancer cells.

Bivalent Genes Targeting of Glioma Heterogeneity and Plasticity

Mariam Markouli et al.

Summary: Gliomas, characterized by high aggressiveness and low survival rates, are mainly attributed to their heterogeneity and complex pathophysiology. Recent data indicate the pivotal role of bivalent genes in regulating gene expression and cell differentiation in glioma pathogenesis, pointing towards their implication in tumor heterogeneity and plasticity. Genes like Homeobox genes, Wingless-Type MMTV Integration Site Family Members, Hedgehog protein, and Solute Carrier Family members control various cellular functions involved in tumor progression and therapy resistance.

Decoding the function of bivalent chromatin in development and cancer

Dhirendra Kumar et al.

Summary: Bivalent chromatin does not rapidly activate genes but protects them from de novo DNA methylation. The activation of bivalent genes in differentiating ESCs is not faster than other silent genes, challenging the instructive role of H3K4me3 in transcription. Loss of H3K4me3 may make bivalent genes more susceptible to aberrant DNA methylation in diseases like cancer.

DNA methylation and breast cancer: A way forward

Maria Teresa Vietri et al.

Summary: This review discusses the DNA methylation changes potentially involved in breast cancer development and their potential clinical utility in diagnosis, prognosis, and therapy. The inactivation of tumor suppressor genes through promoter hypermethylation, detection of targeted and genome-wide DNA methylation changes through liquid-based assays, and the use of epigenetic drugs in combination with anticancer therapies show promise in personalized treatment strategies for breast cancer patients.

SPOP mutation induces DNA methylation via stabilizing GLP/G9a

Jianong Zhang et al.

Summary: The study demonstrates that mutations in the SPOP gene contribute to aberrant DNA hypermethylation in prostate cancer cells, resulting in downregulation of tumor suppressor genes. Treatment with DNA methylation inhibitor can reverse this effect, inhibit tumor cell growth, and enhance the efficacy of chemotherapy drugs.

LSD1 and Aberrant DNA Methylation Mediate Persistence of Enteroendocrine Progenitors That Support BRAF-Mutant Colorectal Cancer

Samuel A. Miller et al.

Summary: In BRAF-mutant colorectal cancer, EEC progenitor cells are enriched and blocked from further differentiation by DNA methylation and NEUROD1 gene silencing. Secretory cells and factors they secrete promote colony formation and cell survival pathways, and can be suppressed by LSD1 inhibition. These findings highlight the important role of EEC progenitors in supporting colorectal cancer progression.

Navigating the DNA methylation landscape of cancer

Atsuya Nishiyama et al.

Summary: DNA methylation is a chemical modification that defines cell type and lineage through controlling gene expression and genome stability. Aberrant DNA methylation in cancer cells targets CpG islands in gene expression regulatory elements. Recent studies suggest that the classical model of DNA hypermethylation in cancer needs to be reevaluated.
