JASPAR 2022: the 9th release of the open-access database of transcription factor binding profiles

Jaime A. Castro-Mondragon et al.

Summary: JASPAR is an open-access database containing manually curated, non-redundant transcription factor binding profiles across six taxonomic groups. The latest release introduces new profiles for plants, vertebrates, urochordates, and insects, as well as new tools for TFBS enrichment analysis and data access. The database can be accessed through various platforms, including the JASPAR website, RESTful API, R/Bioconductor data package, and a new Python package pyJASPAR.

Loss of the Ash2l subunit of histone H3K4 methyltransferase complexes reduces chromatin accessibility at promoters

Mirna Barsoum et al.

Summary: This study investigates the impact of Ash2l loss on gene expression and cellular function. The results indicate that Ash2l loss leads to a decrease in H3K4me3 and changes in chromatin accessibility, affecting gene transcription and cellular function.

H1 histones control the epigenetic landscape by local chromatin compaction

Michael A. Willcockson et al.

Summary: Histone H1 functions as a critical regulator of gene silencing by controlling chromatin compaction, genome organization, and histone methylation. Experimental evidence using a conditional triple-knockout mouse strain demonstrates that H1 plays a key role in regulating the activity of chromatin domains.

Anomalous collapses of Nares Strait ice arches leads to enhanced export of Arctic sea ice

G. W. K. Moore et al.

Summary: Ice arches at the northern and southern ends of Nares Strait, a key passage in the Arctic, are forming for shorter durations, leading to increased ice transport and accelerating the export of multi-year ice.

Mutations and variants of ONECUT1 in diabetes

Anne Philippi et al.

Summary: This study demonstrates that mutations in ONECUT1 can cause monogenic recessive syndromic diabetes and define a subgroup of patients with early-onset diabetes who respond well to treatment. Additionally, variations in ONECUT1 are associated with multifactorial type 2 diabetes. The loss of ONECUT1 impairs pancreatic progenitor formation and endocrine program, highlighting its broad contribution to diabetes pathogenesis.

Transcriptional changes and the role of ONECUT1 in hPSC pancreatic differentiation

Sandra Heller et al.

Summary: The study analyzed transcriptomic and epigenetic datasets from human PSCs differentiating into pancreatic progenitors, revealing the crucial role of the transcription factor ONECUT1 in pancreatic development. ONECUT1 is specifically expressed in pancreatic development and interacts with other key transcription factors during endocrine differentiation. By dissecting stage-specific RNA, ChIP, and ATAC sequencing data, the study provides insights into the regulatory mechanisms of pancreatic development, highlighting ONECUT1 as a key regulator in pancreas progenitors and for endocrine specification.

Integration of feeding behavior by the liver circadian clock reveals network dependency of metabolic rhythms

Carolina M. Greco et al.

Summary: The study investigated the interaction between feeding and the liver clock by reconstituting Bmal1 in hepatocytes, finding that BMAL1 and CEBPB cooperate to regulate daily liver metabolic transcriptional programs. The liver clock and feeding rhythm are sufficient to drive temporal carbohydrate homeostasis, while liver rhythms tied to redox and lipid metabolism require communication with the skeletal muscle clock, highlighting peripheral clock cross-talk.
