D-Alanine Metabolism via D-Ala Aminotransferase by a Marine Gammaproteobacterium, Pseudoalteromonas sp. Strain CF6-2

The prevalence of D-Ala aminotransferase sequences in marine bacteria and metagenomes, particularly in seawater samples, suggests that Gammaproteobacteria is the dominant group utilizing D-Ala for recycling and mineralization in the ocean.

A co-expression vector for baculovirus-mediated protein expression in mammalian cells

Summary: The BacMam system, utilizing baculovirus as a delivery tool, is widely used for recombinant production of eukaryotic proteins in mammalian cells. In this study, a BacMam vector (pBMCL1) was developed, which allows easy tracking of virus production, enhances infection efficiency in mammalian cells, reduces unwanted transcription of toxic genes in insect cells, and enables the co-expression of multiple proteins using a single virus. The successful application of the pBMCL1 vector in expressing homo-tetrameric human TRPC3 channel and hetero-octameric KATP channel is demonstrated.

This system shows potential for future industrial applications and offers an alternative to previous plasmid-carrying E. coli expression systems.

In a study of Thermotoga maritima, a bacterium lacking a broad-spectrum racemase, it was found that the TM0831 enzyme is capable of catalyzing both D-amino acid aminotransferase reaction and amino acid racemase activity to produce D-glutamate.

Summary: D-amino acids are valuable building blocks for the synthesis of biologically active compounds and pharmaceuticals. The efficient asymmetric synthesis of optically pure D-amino acids using a pyridoxal-5'-phosphate-dependent transaminase from Haliscomenobacter hydrossis was investigated. A one-pot three-enzyme system was constructed, achieving high product yield and enantiomeric excess. Factors such as high concentration of alpha-keto acids and low reaction temperature were found to reduce cofactor leakage and increase stability of the transaminase.
