Summary: Terpenoids, as the largest family of natural products, play significant roles in medicine, agriculture, cosmetics, and food. Traditional extraction methods have low yields, waste resources, and are not suitable for endangered species. Modern biotech approaches are promising in discovering and characterizing plant terpenoid-related P450 enzymes, and metabolic engineering strategies can be employed to increase terpenoid production.

Studies involving crystal structures of IPK variants from Candidatus methanomethylophilus alvus (CMA) reveal substrate promiscuity towards non-natural substrates and shed light on the role of active-site residues in catalysis. Active-site engineering and mutation studies further confirm the importance of conserved motifs in catalysis, providing insights for the development of IPKs as useful biocatalysts.

Engineering Escherichia coli for effective and economic production of cis-abienol by optimizing isopentenol utilization pathway

By screening and coordinating alcohol kinases and isopentenyl phosphate kinases (IPKs), the cis-abienol yield was significantly improved. By combining enzyme engineering and flux control using the non-canonical isopentenol utilization (IU) pathway, high purity production of linalool was achieved, paving the way for efficient optimization of terpenoid production in engineered microbes.
