Palladium-catalyzed direct γ-C(sp3)-H arylation of β-alkoxy cyclohexenones: reaction scope and mechanistic insights

Li-Dong Shao et al.

Summary: A mild Pd-catalyzed method for direct gamma-C(sp(3))-arylation of unactivated beta-alkoxy cyclohexenones is reported. The method is well tolerated with ortho-hetero substituted aryl bromides and sterically hindered ortho-dimethoxy or rigid ortho-diaryl aryl bromides, showing excellent gamma-site selectivity. The method enables the facile synthesis of CBD-like compounds with selective anti-proliferative effects against glioma stem cells.

Anti-Adipogenic Lanostane-Type Triterpenoids from the Edible and Medicinal Mushroom Ganoderma applanatum

Xing-Rong Peng et al.

Summary: This study focused on the lanostane triterpenoids from Ganoderma applanatum and isolated forty highly oxygenated compounds, including sixteen new compounds. The structures of these compounds were identified and their anti-adipogenesis activities were evaluated. Some compounds exhibited stronger effects than the positive control and a key role of the A-seco-23 -> 26 lactone skeleton in anti-adipogenesis activity was established.

Applanoids A-E as the First Examples of C-15/C-20 Michael Adducts in Ganoderma Triterpenoids and Their PXR Agonistic Activity

Hai-Guo Su et al.

Summary: In this study, nine new Ganoderma triterpenoids (GTs) were isolated from the medicinal fungus Ganoderma applanatum, and their structures and absolute configurations were determined. Compounds 1-8 were evaluated for their hPXR agonistic activity, and compounds 1, 2, and 4 showed dose-dependent activation of hPXR. This study further expands our knowledge of the structural diversity of GTs and provides insights for the discovery of PXR agonists from GTs.

Retromer regulates the lysosomal clearance of MAPT/tau

Julian M. Carosi et al.

Summary: The autophagy-lysosome axis is crucial for the clearance of MAPT aggregates, with dysfunction of the central retromer component VPS35 leading to impaired autophagic resolution and accumulation of cytoplasmic MAPT aggregates. Maintaining the integrity of the retromer complex regulates the autophagy-lysosome axis to suppress MAPT aggregation and propagation.

Ganoderic Acid A To Alleviate Neuroinflammation of Alzheimer's Disease in Mice by Regulating the Imbalance of the Th17/Tregs Axis

Yan Zhang et al.

Summary: This study found that Ganoderic acid A can alleviate neuroinflammation in AD mice by regulating the imbalance of the Th17/Tregs axis. The mechanism may be related to inhibition of the JAK/STAT signaling pathway induced by Th17 cells and enhancement of mitochondrial oxidative phosphorylation by regulating Treg cells, improving mitochondrial dysfunction in AD mice.
