Summary: Two novel CLIC1 blockers, Q48 and Q54, were identified as more potent antiproliferative agents for GSCs than metformin in vitro. Q48 and Q54 also showed inhibitory effects on GSC self-renewal, migration and invasion, with low systemic toxicity in vivo. These findings suggest that Q48 and Q54 may be potential CLIC1 blockers for the treatment of CLIC1-expressing glioblastomas.

Summary: This study investigated the blood-brain barrier permeability of Skepinone-based p38 alpha MAP kinase inhibitors and their potential therapeutic effects in neuroinflammation and neurodegeneration. Methyl ester prodrugs were used to enhance oral absorption, resulting in compounds with good brain-to-plasma ratios. These compounds were selective for p38 alpha-MAP kinase, metabolically stable, and able to modulate IL-6 and TNF-alpha production.

Design, synthesis and evaluation of novel scutellarin and scutellarein-N, N-bis-substituted carbamate-L-amino acid derivatives as potential multifunctional therapeutics for Alzheimer's disease

Summary: In this study, a series of scutellarin derivatives were designed, synthesized, and evaluated as multifunctional therapeutic agents for Alzheimer's disease. One of the compounds, 6h, showed potent inhibition of acetyl cholinesterase and Aβ aggregation, as well as neuroprotective effects and optimal blood-brain barrier and intestinal absorption characteristics. The in vivo experiments further demonstrated its potential in improving learning and memory impairment. These findings highlight compound 6h as a promising multifunctional drug candidate against Alzheimer's disease.

Discovery of a New CaMKII-Targeted Synthetic Lethal Therapy against Glioblastoma Stem-like Cells

Summary: The poor prognoses of patients with glioblastoma multiforme (GBM) are attributed to glioblastoma stem-like cells (GSCs) which induce drug resistance and rapid tumor recurrence. This study aims to explore a novel synthetic lethal therapy targeting calcium/calmodulin-dependent protein kinase II (CaMKII) for combatting GSCs. Through high-throughput drug combination screening, the combined treatment of CaMKII inhibitors and neurokinin 1 receptor (NK1R) inhibitors is found to exhibit chemical synthetic lethal effects on GSCs both in vitro and in vivo. It is proposed that this CaMKII-targeted combination therapy could be effective for treating GBM.

Chalcones as Anti-Glioblastoma Stem Cell Agent Alone or as Nanoparticle Formulation Using Carbon Dots as Nanocarrier

Summary: The current prognosis for glioblastoma is poor, largely due to treatment-resistant glioblastoma stem cells and the limited ability of drugs to reach therapeutic levels within the tumor. However, chalcones, aromatic ketones, have shown promise in reducing malignant properties in cancers, including glioblastoma. Nanomedicines, such as carbon-dots, can enhance drug accumulation and induce tumor cell death. In this study, a series of 4'-amino chalcones were synthesized and tested for their cytotoxicity against glioblastoma stem cells. Some of the chalcones exhibited significant cytotoxicity, and when attached to transferrin-conjugated carbon-dots, their cytotoxicity was further enhanced. These findings suggest that certain chalcones may serve as lead compounds for the development of novel anti-glioblastoma stem cell drugs, and carbon-dot mediated drug delivery is a promising approach.

An Integrated Bioinformatics Study of a Novel Niclosamide Derivative, NSC765689, a Potential GSK3 beta/beta-Catenin/STAT3/CD44 Suppressor with Anti-Glioblastoma Properties

Summary: Despite current standard treatment options, including surgery, radiation, and chemotherapy, glioblastoma multiform (GBM) remains resistant to therapy due to the presence of glioma stem cells (GSCs). In this study, a newly synthesized compound, NSC765689, was identified as a potential pharmacological lead compound for targeting GBM oncogenic signaling pathways. Through in silico molecular docking and in vitro studies, NSC765689 showed promising anti-proliferative and cytotoxic effects against GBM cells by targeting key oncogenes such as GSK3 beta, beta-Catenin, STAT3, and CD44, as well as upregulating the tumor suppressor miR-135b. Further research is needed to validate the efficacy of NSC765689 as a novel potential therapeutic for GBM.
