Integrative functional linear model for genome-wide association studies with multiple traits

Yang Li et al.

Summary: The integrative functional linear model proposed in this study effectively addresses the challenges posed by high-dimensional SNP data and correlations among multiple traits in GWAS. It demonstrates satisfactory performance in the identification of disease-associated genetic variants and provides biologically meaningful findings in the analysis of type 2 diabetes data.

Sparse functional linear discriminant analysis

Juhyun Park et al.

Summary: Functional linear discriminant analysis is a simple yet efficient method for classification that can achieve perfect classification. This paper proposes a new approach that incorporates sparsity into the functional setting, resulting in a solution that is easier to interpret without compromising performance. Using simulated and real data examples, the method is shown to consistently estimate the true function, regardless of whether or not it is sparse.

Truncated estimation in functional generalized linear regression models

Xi Liu et al.

Summary: Functional generalized linear models investigate the effect of functional predictors on a scalar response, with a focus on the influence of functional predictors' values up to a certain point on the conditional mean. This paper proposes a penalized likelihood estimator that combines structured variable selection and localized B-spline expansion to estimate the regression coefficient function. The method includes a nested group lasso penalty to ensure desired truncation on the estimator, and an optimization scheme for efficient computation.

Gene-level association analysis of ordinal traits with functional ordinal logistic regressions

Chi-Yang Chiu et al.

Summary: In this paper, functional ordinal logistic regression (FOLR) models are proposed for gene-based analysis of ordinal traits. The models are built upon functional data analysis and can effectively handle common and rare genetic variant data, achieving direct analysis of ordinal traits with good performance. In the application to Age-Related Eye Disease Study data, FOLR models successfully identified two genes strongly associated with four ordinal traits.

Gene-Based Association Testing of Dichotomous Traits With Generalized Functional Linear Mixed Models Using Extended Pedigrees: Applications to Age-Related Macular Degeneration

Yingda Jiang et al.

Summary: Genetics plays a significant role in age-related macular degeneration (AMD) and the study utilized a new generalized functional linear mixed model (GFLMM) to test for gene-based association in AMD families. Significant associations were found with known AMD genes CFH and ARMS2, while suggestive signals were observed in genes like ASAH1, CLEC6A, TMEM63C, and SGSM1. The GFLMM-based statistics provided a new tool for conducting family-based genetic studies of complex diseases.

An association test for functional data based on Kendall's Tau

Sneha Jadhav et al.

Summary: This paper addresses the issue of testing the association between a functional variable in a Hilbert space and a scalar variable. A distribution-free test statistic based on Kendall's Tau is proposed, and the distribution under the null hypothesis of independence is established using the theory of U-statistics. The method is shown to outperform alternatives in multiple settings through simulations, highlighting its effectiveness and robustness.

Identifying Gene-Environment Interactions With Robust Marginal Bayesian Variable Selection

Xi Lu et al.

Summary: A novel marginal Bayesian variable selection method for GxE studies is proposed in this study, which is robust to data contamination and outliers in the outcome variables. The method has outperformed several alternatives in extensive simulation studies and has been further demonstrated in case studies using data from the Nurse Health Study (NHS), showing important biological implications of the identified main and interaction effects from the real data analysis.
