Engineering the multiscale complexity of vascular networks

Colleen O'Connor et al.

Summary: This Review discusses how materials advances have facilitated the development of vascular engineering approaches, driven by both technology and nature, to create vascular networks for various applications. It emphasizes the importance of understanding vascular structure and highlights new methods such as bioprinting, organoids and microfluidic systems for fabricating 3D vascular topologies at a cellular scale with lumen perfusion. The Review also highlights the remaining knowledge gaps and opportunities in this field.

A modular polymer microbead angiogenesis scaffold to characterize the effects of adhesion ligand density on angiogenic sprouting

Marianna Sofman et al.

Summary: Three-dimensional micro-physiological in vitro models, such as embedding degradable hydrogel microbeads coated with induced pluripotent stem cell-derived endothelial cells, have the potential to drive angiogenesis and support vascularization of cell-dense tissues with high metabolic demand.

Cell-Laden Gradient Hydrogel Scaffolds for Neovascularization of Engineered Tissues

Yusheng J. He et al.

Summary: This study utilized a photopolymerization approach to create gradient scaffolds for spatial encapsulation of vascular spheroids, revealing that scaffolds with immobilized RGD gradients promoted biased vascular sprouting towards higher RGD concentrations. Conversely, combined gradient scaffolds with opposing gradients in RGD and modulus reduced biased sprouting responses.
