Summary: Sacrificial printing involves using 3D-printed sacrificial ink to create open-channels in hydrogel constructs. Traditional sacrificial inks lack the ability to mimic tissue development dynamics. To overcome this, a new class of sacrificial inks was developed with tailorable and programmable delayed dissolution profiles. These inks showed compatibility with various biofabrication technologies and allowed precise control over the introduction of architectural features into cell-laden hydrogel constructs.

Species-Based Differences in Mechanical Properties, Cytocompatibility, and Printability of Methacrylated Collagen Hydrogels

Summary: Collagen methacrylation is a promising method for generating cell-laden hydrogels with improved mechanical properties. In this study, the effects of collagen species, collagen extraction methods, and photoinitiators on the physical properties of methacrylated collagen hydrogels were investigated. Human collagen showed the highest methacrylation degree, and lithuim phenyl-2,4,6-trimethylbenzoylphosphinate was more cytocompatible than Irgacure-2959. The rat methacrylated collagen hydrogels had higher compressive modulus and cell viability compared to bovine collagen hydrogels. Human methacrylated collagen hydrogels demonstrated superior print fidelity.

Characterization of Tuna Gelatin-Based Hydrogels as a Matrix for Drug Delivery

Summary: The skin of yellowfin tuna contains abundant collagen, which can be hydrolyzed to generate gelatin suitable for forming hydrogels. Novel tuna gelatin hydrogels were designed as drug vehicles for local administration after surgical resection. Characterization and morphological studies confirmed the heterogeneity composition and porous network structure of the gelatin. The properties and transition of the hydrogels were investigated using dynamic rheology and differential scanning calorimetry. Drug loading and release experiments demonstrated sustained release of the drugs from the gelatin hydrogels, highlighting the importance of material composition in drug delivery.

Designing Gelatin Methacryloyl (GelMA)-Based Bioinks for Visible Light Stereolithographic 3D Biofabrication

Summary: Optimized synthesis parameters were used to develop GelMA bioinks with slow sol-gel transition at room temperature and visible light crosslinkable functions. Eight GelMA combinations suitable for DLP-SLA bioprinting were identified through systematic physical and rheological property characterizations. RO-GelMA bioinks were found to be most suitable for achieving high resolution in DLP-SLA printing, showing excellent biocompatibility and cell survival rates.
