Impact of Cell Loading of Recombinant Spider Silk Based Bioinks on Gelation and Printability

Annika Lechner et al.

Summary: The printability of bioinks refers to considerations such as rheology, extrudability, filament formation, shape fidelity, cell viability, and cellular development post-printing. Recent studies indicate that recombinant spider silk hydrogels show high shape fidelity and may be suitable for bioink formulations, with the encapsulation of cells impacting gelation and mechanics differently depending on the silk variant used. RGD-modified spider silk hydrogels are physically crosslinked by cells, showing high cell viability after extrusion-based printing.

3D Bioprinting of Multifunctional Dynamic Nanocomposite Bioinks Incorporating Cu-Doped Mesoporous Bioactive Glass Nanoparticles for Bone Tissue Engineering

Hui Zhu et al.

Summary: Bioprinting has made significant progress in recent years in the fabrication of complex bionic tissues. However, developing cell-laden bioinks with superior physiochemical properties and bio-functionality remains challenging. In this study, a multifunctional nanocomposite bioink was developed using amine-functionalized copper-doped mesoporous bioactive glass nanoparticles and a hydrogel formulation based on dynamic covalent chemistry. This bioink showed favorable rheological properties, improved shape fidelity, and structural stability for extrusion-based bioprinting. The introduction of aminated particles and the reversible dynamic microenvironment enabled rapid spreading and high survival of embedded cells. The bioprinted scaffolds promoted osteogenic differentiation and angiogenesis of cells without additional growth factors in vitro. These nanocomposite bioinks have great potential for bioprinting complex 3D environments in bone tissue engineering.

Calcium supplementation of bioinks reduces shear stress-induced cell damage during bioprinting

Lena Fischer et al.

Summary: During bioprinting, cells are subjected to shear stress, which can cause cell damage and death. Monitoring and controlling shear stress-induced cell damage is important but currently challenging. This study shows that cell deformation, FM 1-43 incorporation, and cell death are influenced by cell stiffness. Calcium supplementation reduces shear stress-induced cell damage and death, but not cell deformation.
