Continuous millisecond conformational cycle of a DEAH box helicase reveals control of domain motions by atomic-scale transitions

Summary: Helicases are motor enzymes that play important roles in maintaining genome stability and controlling recombination. Using molecular dynamics simulations, this study reveals the atomic-level conformational cycle of RNA translocation by the DEAH-box helicase Prp43. The findings highlight the complex atomic-scale transitions involved in the crawling movement of the helicase along RNA.

Regulation of the DEAH/RHA helicase Prp43 by the G-patch factor Pfa1

Summary: The DEAH/RHA helicase Prp43 plays an important role in RNA splicing and ribosome biogenesis. Using single-molecule Forster Resonance Energy Transfer (smFRET) technology, we discovered that the RecA domains in Prp43 undergo conformational changes during catalysis, and Pfa1(gp) can induce alternating between open and closed states. Additionally, Pfa1(gp) enables Prp43 to switch between RNA-bound and RNA-unbound states, instead of dissociating from the RNA.

Kinetic and structural mechanism for DNA unwinding by a non-hexameric helicase

Summary: The study utilized optical tweezers to directly measure the stepping behavior of UvrD as it processes a DNA hairpin, revealing a variable step size averaging around 3 base pairs. Analysis of ATP hydrolysis kinetics showed that UvrD moves one base pair at a time but non-uniformly releases nascent single strands, suggesting a conserved mechanism among other non-hexameric helicases.
