Ultrasonic-assisted extraction of polysaccharides from coix seeds: Optimization, purification, and in vitro digestibility

Summary: The study optimized the extraction of polysaccharides from coix seeds using response surface methodology with ultrasound as an auxiliary method. The results showed that ultrasound extraction led to higher polysaccharide content and lower molecular weight compared to hot water extraction. Summary: A new PMA-qPCR method was developed in this study for the rapid and accurate detection of viable bacteria and spores of marine-derived Bacillus velezensis B-9987. Compared to the traditional qPCR method, the PMA-qPCR method showed higher sensitivity in detecting viable bacteria in mixed samples. In pure culture conditions, this method could accurately and quickly detect viable B-9987 bacteria and spores in marine Bacillus wettable powder within 3-4 hours.

Hydrophobic magnetic deep eutectic solvent: Synthesis, properties, and application in DNA separation

Summary: In this study, four hydrophobic magnetic deep eutectic solvents (HMDESs) were designed and prepared for DNA extraction, with the conformations of HMDESs simulated and their H-bonding interactions investigated using density functional theory (DFT) calculations. The physical and thermal properties of HMDESs were characterized, and DNA extracted by HMDESs could be quickly collected by an external magnet. Three auxiliary extraction methods were introduced, and factors impacting the DNA extraction process were systematically investigated through single-factor experimental analysis. The proposed extraction method successfully extracted DNA from complex matrices and E. coli cell lysate, and the binding sites between HMDESs and DNA were studied.

Isolation, stability, and characteristics of high-pressure superdormant Bacillus subtilis spores

Summary: Bacterial spores pose a challenge in industrial decontamination due to their extreme resistance, but a combination of high pressure and temperature can trigger their germination. However, a subpopulation of high-pressure superdormant (HPSD) spores hinders the implementation of this strategy. Factors influencing the prevalence of HPSD spores were investigated, showing a dependency on pressure treatment times and the absence of major germinant receptors leading to increased HPSD spores. Efforts to understand the properties and underlying causes of HPSD spores are crucial for developing effective germination-inactivation strategies involving high pressure.
