Manipulate tumor hypoxia for improved photodynamic therapy using nanomaterials

Mengqi Yi et al.

Summary: Photodynamic therapy (PDT) has gained significant interest due to its low adverse effects, minimal invasiveness, and high patient compliance. However, tumor hypoxia significantly hampers the therapeutic effect of PDT, leading to tumor cell proliferation, invasion, and metastasis. Recent investigations have focused on using nanomaterials to overcome tumor hypoxia and enhance PDT efficacy.

Dually enhanced phototherapy by gambogic acid and hyperthemia-activated chemotherapy for synergistic breast cancer treatment

Yuhan Wang et al.

Summary: Combining chemotherapy and phototherapy via nanoscale drug delivery systems has been explored to improve cancer treatment. However, excessive antioxidants and heat shock proteins in tumor cells hinder the efficacy of phototherapy. To address these challenges, researchers developed a novel phase change material-based nanoplatform that can release chemotherapy drugs and enhance phototherapy by regulating reactive oxygen species levels and suppressing heat resistance proteins.

Guiding Transition Metal-Doped Hollow Cerium Tandem Nanozymes with Elaborately Regulated Multi-Enzymatic Activities for Intensive Chemodynamic Therapy

Shuming Dong et al.

Summary: The authors developed a hollow mesoporous Mn/Zr-co-doped CeO2 tandem nanozyme (PHMZCO-AT) with regulated multi-enzymatic activities to disrupt the H2O2 homeostasis, achieve intensive NCDT, and induce tumor apoptosis and death. The nanozyme catalyzes the conversion of superoxide anion into H2O2 in the tumor region and inhibits the off-target decomposition of H2O2. The elevated H2O2 is then converted into toxic hydroxyl radicals, leading to tumor cell death.

Tumor-specific and photothermal-augmented chemodynamic therapy by ferrocene-carbon dot-crosslinked nanoparticles

Shan Sun et al.

Summary: Extensive research has been carried out on multifunctional theranostic agents for cancer treatment. In this study, ferrocene-carbon dot-crosslinked nanoparticles were designed and fabricated for specific and enhanced chemotherapy. These nanoparticles not only inherit the properties of carbon dots but also demonstrate chemodynamic therapy selectively in tumor microenvironment due to the presence of ferrocene. The enlarged particle size of the nanoparticles facilitates their accumulation at tumor sites and results in improved antitumor outcomes. The combination of photothermal therapy and chemodynamic therapy shows a synergistic therapeutic effect. This study provides a facile approach to fabricate multifunctional theranostic nanoplatforms and emphasizes the benefits of synergizing therapeutic modalities for cancer therapy.
