Fluorination Promotes the Cytosolic Delivery of Genes, Proteins, and Peptides

Jia Lv et al.

Summary: This study proposes the use of fluorous ligands to promote cytosolic delivery of biomolecules. Fluorination significantly improves gene delivery efficacy and serum tolerance of cationic polymers. Fluorination also enhances multiple steps such as cellular uptake and endosomal escape, resulting in efficient gene delivery and minimal material toxicity. Additionally, fluorinated polymers are efficient for intracellular protein delivery and cytosolic peptide delivery, outperforming traditional techniques.

Enhanced transfection efficiency of low generation PAMAM dendrimer conjugated with the nuclear localization signal peptide derived from herpesviridae

Jeil Lee et al.

Summary: PAMAM dendrimer, a widely studied polymer in biomedical research, shows potential as an alternative to PEI in gene delivery when conjugated with NLS sequences. This study suggests that PAMAM generation 2 with NLS sequences can reduce cytotoxicity while maintaining high transfection efficiency.

Non-viral Vectors in Gene Therapy: Recent Development, Challenges, and Prospects

Hui Zu et al.

Summary: Although non-viral vectors in gene therapy have low cytotoxicity and immunogenicity, they still face challenges in gene transfer efficiency, specificity, gene expression duration, and safety. Overcoming these obstacles is necessary to enhance the effectiveness of non-viral vectors in gene therapy.
