A Window to the Potassium World. The Evidence of Potassium Energetics in the Mitochondria and Identity of the Mitochondrial ATP-Dependent K+ Channel

Dmitry B. Zorov

Summary: The conclusions made by Juhaszova et al. in their three papers published in Function are considered a breakthrough in bioenergetics and mitochondrial medicine. They discovered that ATP synthase not only transports protons, but also potassium ions, leading to the generation of ATP. This potassium ion transport not only contributes to energy production, but may also play a positive role in protective anti-ischemic signaling.

The Effect of Uridine on the State of Skeletal Muscles and the Functioning of Mitochondria in Duchenne Dystrophy

Mikhail Dubinin et al.

Summary: Duchenne muscular dystrophy is a disease caused by the loss of dystrophin, resulting in metabolic impairment and muscle weakness. This study investigated the potential therapeutic effects of uridine on the skeletal muscles of mdx mice, a model for Duchenne muscular dystrophy. The results showed that uridine administration reduced fibrosis in the muscles, but had no effect on degeneration/regeneration cycles, inflammation, pseudohypertrophy, or muscle strength. Uridine also had no significant impact on mitochondrial function and OXPHOS dysfunction in mdx mouse muscles. However, it increased the expression of Drp1 and Parkin genes, suggesting a potential improvement in organelle fission and mitophagy. Uridine also restored potassium transport and reduced reactive oxygen species production, but did not affect calcium retention capacity in mdx mouse mitochondria.

ATP Synthase K+- and H+-Fluxes Drive ATP Synthesis and Enable Mitochondrial K+-Uniporter Function: I. Characterization of Ion Fluxes

Magdalena Juhaszova et al.

Summary: ATP synthase can utilize both Delta psi(m)-driven H+- and K+-transport to synthesize ATP under physiological conditions. The presence of K+ increases ATP synthesis rate in isolated mitochondria while also increasing oxygen consumption rate. The data obtained from purified F1Fo single molecule experiments are consistent with the functional data observed in intact mitochondria.

ATP Synthase K+- and H+-fluxes Drive ATP Synthesis and Enable Mitochondrial K+-Uniporter Function: II. Ion and ATP Synthase Flux Regulation

Magdalena Juhaszova et al.

Summary: ATP synthase serves as the primary way for K+ to enter mitochondria, and its activity can be upregulated by endogenous survival-related proteins via IF1. The interaction between IF1 and Bcl-xL or Mcl-1 enhances ATP synthase's efficiency and limits ischemia-reperfusion injury.

Mikhail Dubinin et al.

Summary: The study synthesized pyridinium derivatives of betulin, showing their effects on artificial membrane systems and cytotoxicity in cell models. The presence of methyl groups in the compounds influenced their ability to permeabilize membranes and affect mitochondrial function. The study highlights the dependence of membranotropic and biological actions on the structure and lipophilicity of the derivatives.

Metformin Reverses the Enhanced Myocardial SR/ER-Mitochondria Interaction and Impaired Complex I-Driven Respiration in Dystrophin-Deficient Mice

Claire Angebault et al.

Summary: In the early stages of Duchenne muscular dystrophy (DMD), the heart exhibits impaired sarco-endoplasmic reticulum (SR/ER)-mitochondria interactions and mitochondrial dysfunction, resulting in disrupted mitochondrial Ca2+ homeostasis and complex I dysfunction. Treatment with metformin could reverse these abnormalities, offering a potential therapeutic perspective in DMD.

Mitochondrial K+ channels and their implications for disease mechanisms

Vanessa Checchetto et al.

Summary: The field of mitochondrial ion channels has rapidly developed in the past decade due to molecular identification and specific pharmacological targeting. Despite challenges in genetic manipulation of mitochondrial K+ channels, they are recognized to play important roles in various pathologies and are druggable targets for potential therapeutic interventions. The in-depth investigation of mitochondrial potassium channels may lead to the development of innovative small molecules with significant therapeutic potential.

BK in Double-Membrane Organelles: A Biophysical, Pharmacological, and Functional Survey

Summary: This article introduces the discovery and detailed research on calcium-activated potassium currents and the Ca2+-activated potassium channel, BK. The BK channel, with its modular structure and distribution in different cell types, is regulated by Ca2+, voltage, and auxiliary subunits. While the precise roles of mitoBK and nBK are still largely unknown, mitoBK has been reported to play a protective role in heart ischemic injury, and nBK may be involved in regulating nuclear Ca2+, membrane potential, and eNOS expression.

Abnormal Calcium Handling in Duchenne Muscular Dystrophy: Mechanisms and Potential Therapies

Satvik Mareedu et al.

Summary: Duchenne muscular dystrophy is a muscle-wasting disease caused by the loss of dystrophin, leading to muscle degeneration, necrosis, inflammation, and eventual weakness and premature death. Abnormal elevation of intracellular calcium in the dystrophin-deficient muscle is a major contributing factor to disease progression. Targeting calcium-handling proteins and mechanisms could be a promising therapeutic strategy for DMD.
