Oleoylethanolamide attenuates the stress-mediated potentiation of rewarding properties of cocaine associated with an increased TLR4 proinflammatory response

Macarena Gonzalez-Portilla et al.

Summary: The messenger molecule OEA has been found to have diverse physiological functions including metabolism, immune response, and now also affects reward-related behavior. Social stress has been shown to increase cocaine sensitivity, but pretreatment with OEA can prevent this effect and exert an anti-inflammatory action in the brain. These findings suggest that OEA may have a protective effect against stress-induced drug addiction.

CB1 receptor antagonist AM4113 reverts the effects of cannabidiol on cue and stress-induced reinstatement of cocaine-seeking behaviour in mice

Miguel A. Lujan et al.

Summary: The study found that cannabidiol has complex effects on the behavioral performance of rodent models of cocaine addiction, including no effect on extinction learning, increase in CB1 receptor protein levels, reduction in FosB accumulation, increase in GluR2 AMPA receptor subunits, attenuation of cue-induced reinstatement of drug seeking, but facilitation of stress-induced restoration of drug seeking. These findings suggest that cannabidiol induces a variety of CB1-related changes that have varying impacts on cocaine-seeking behavior, potentially limiting its therapeutic applications.

Oleoylethanolamide attenuates the stress-mediated potentiation of rewarding properties of cocaine associated with an increased TLR4 proinflammatory response

Macarena Gonzalez-Portilla et al.

Summary: The lipid-derived messenger oleoylethanolamide (OEA) plays a role in regulating metabolism and the immune response. Recent research has shown that OEA also affects reward-related behavior. This study found that OEA pretreatment can prevent the increased sensitivity to cocaine induced by social defeat stress, and this protective effect may be mediated by its anti-inflammatory action on Toll-like receptor 4 (TLR4) in the cerebellum.
