Pharmacokinetics and Toxicities of Oral Docetaxel Formulations Co-Administered with Ritonavir in Phase I Trials

Summary: Severe toxicity was observed in 25% of patients treated with oral docetaxel in combination with ritonavir, with mucositis, fatigue, diarrhea, nausea, and vomiting being the most frequent adverse events. The severe toxicity group had significantly higher docetaxel AUC and C-max compared to the non-severe toxicity group. Future randomized trials are needed to further evaluate the toxicity and efficacy of the new weekly oral docetaxel and ritonavir regimen in comparison to standard IV docetaxel.
