Sestrin2 induction contributes to anti-inflammatory responses and cell survival by globular adiponectin in macrophages

Sumin Lee et al.

Summary: The study demonstrates that in macrophages, globular adiponectin modulates cell survival and anti-inflammatory responses by activating the sestrin2 signaling pathway. The induction of sestrin2 is crucial for autophagy activation and enhanced cell viability by adiponectin.

Blockade of Autophagy Prevents the Progression of Hyperuricemic Nephropathy Through Inhibiting NLRP3 Inflammasome-Mediated Pyroptosis

Yan Hu et al.

Summary: Hyperuricemia, a common metabolic disease, is closely related to various diseases including chronic kidney disease. This study found that persistent uric acid stimulation activated autophagy and facilitated renal fibrosis in rats with hyperuricemic nephropathy. Inhibition of autophagy prevented the upregulation of markers related to fibrosis in human renal tubular cells exposed to uric acid. Furthermore, uric acid promoted autophagy via the p53 pathway. Inhibiting autophagy also alleviated NLRP3 inflammasome activation and pyroptosis in vivo and in vitro. Mechanistically, the release of cathepsin B resulting from elevated autophagy and degradation of autophagolysosomes was found to be involved in NLRP3 inflammasome activation. In summary, inhibition of autophagy may be a promising therapeutic strategy for hyperuricemic nephropathy by preventing fibrosis and NLRP3 inflammasome-mediated pyroptosis.

Autophagy activation and SREBP-1 induction contribute to fatty acid metabolic reprogramming by leptin in breast cancer cells

Duc-Vinh Pham et al.

Summary: Leptin induces growth of breast cancer cells through reprogramming of lipid metabolism and activation of autophagy, with crucial roles played by SREBP-1 induction and fatty acid synthase induction. Autophagy mediates the metabolic effects of leptin on cancer cell-specific metabolism, highlighting its pivotal contribution to tumor growth.
