Evaluation of the immunoprotection efficacy of Riemerella anatipestifer fur-deficient mutant as an attenuated vaccine

Summary: This study demonstrates that the RA CH-1 Afur strain is significantly attenuated and provides effective protection against high-dose RA CH-1 infection in ducklings. Inoculation with the mutant strain did not cause clinical symptoms or significant histological lesions, and the bacteria were cleared by the host within 3 days. Furthermore, RA CH-1 Afur induced high levels of IgY antibodies, which persisted for at least 49 days.

Summary: Iron is essential for bacteria, including the pathogen Riemerella anatipestifer, which utilizes hemin from duck hemoglobin as an iron source. The RhuR-RhuA locus in R. anatipestifer CH-1 is involved in hemin acquisition from duck hemoglobin through a unique pathway. RhuA is an outer membrane hemin-binding protein, while RhuR is a TonB2-dependent hemin transporter, and their functions are interdependent. The homologues of RhuR and RhuA are widely distributed across bacterial species, representing a novel hemin transportation system in Gram-negative bacteria.

Summary: The study identified 26 new variants of the tigecycline resistance gene tet(X) from the poultry pathogen Riemerella anatipestifer, expanding the current family extensively. R. anatipestifer is shown to be a natural reservoir for tet(X), with varied copies of tet(X) progenitors on its chromosome. A potential progenitor of tet(X), Tet(X2/4)-P, was characterized to show intermediate tigecycline resistance.
