β1 Integrin induces adhesion and migration of human Th17 cells via Pyk2-dependent activation of P2X4 receptor

Chakib Hamoudi et al.

Summary: In this study, we found that integrin-mediated signaling induces the release of ATP, promoting T-cell adhesion and migration across fibronectin. P2X4 receptor plays a key role in T-cell adhesion and migration, while P2X7 and P2Y(11) receptors have no effect. Furthermore, we discovered a positive feedback loop between beta 1 integrin and P2X4 receptor signaling pathways.

Identification of a shared genetic risk locus for Kawasaki disease and immunoglobulin A vasculitis by a cross-phenotype meta-analysis

Elio G. Carmona et al.

Summary: This study aimed to increase knowledge on the genetic contribution to Kawasaki disease (KD) and IgA vasculitis (IgAV) by performing a comprehensive large-scale analysis on the genetic overlap between them. A new risk locus with pleiotropic effects on the two childhood vasculitides analyzed was identified, representing the strongest non-HLA signal described for IgAV to date.

CircZDBF2 up-regulates RNF145 by ceRNA model and recruits CEBPB to accelerate oral squamous cell carcinoma progression via NF kappa B signaling pathway

Liang Rong et al.

Summary: This study found that circZDBF2 is highly expressed in OSCC cells and promotes OSCC progression through multiple molecular mechanisms involving miR-362-5p, miR-500b-5p, RNF145, CEBPB, and the NF kappa B signaling pathway.

Identification of susceptibility loci for Takayasu arteritis through a large multi-ancestral genome-wide association study

Lourdes Ortiz-Fernandez et al.

Summary: A genetic study on Takayasu arteritis identified HLA risk factors and several susceptibility loci, shedding light on potential disease mechanisms. Comparison to other traits revealed genetic relatedness to inflammatory bowel disease, with epigenetic patterns suggesting roles for monocytes and B cells in Takayasu arteritis.

Summary: Systemic vasculitides are a type of multisystem blood vessel disorders classified by vessel size. Prompt identification is crucial due to the potentially serious outcomes associated with these conditions. Ethnic and regional differences in incidence and clinical characteristics should be taken into account.

Deficiency in the frequency and function of Tr1 cells in IgAV and the possible role of IL-27

Lu Pan et al.

Summary: Tr1 cells are involved in the pathogenesis of IgAV, with a decreased frequency in peripheral blood but increased in kidney tissue. The impaired suppressive function of Tr1 cells in IgAV is associated with low IL-27 levels and potentially involves the specific transcription factor EGR2. Tr1 cells may be a risk factor for IgAV recurrence.

Genetic Association of a Gain-of-Function IFNGR1 Polymorphism and the Intergenic Region LNCAROD/DKK1 With Behcet's Disease

Lourdes Ortiz Fernandez et al.

Summary: This study identified novel functional variants associated with Behcet's disease and replicated and extended genetic associations to other loci across multiple ancestries. Additionally, over 30 genetic susceptibility loci with a suggestive level of association were identified, requiring further replication. Functional annotation of genetic susceptibility loci revealed potential regulatory roles and suggested causal genes and molecular mechanisms for further investigation.

An open approach to systematically prioritize causal variants and genes at all published human GWAS trait-associated loci

Edward Mountjoy et al.

Summary: In this study, an open resource was introduced to systematically fine map and prioritize genes across published GWAS loci, aiming at identifying causal genes associated with complex traits. By training a machine learning model using genetics and functional genomics data, the authors were able to distinguish causal genes from neighboring genes, outperforming distance-based models. The prioritized genes were found to be enriched for known approved drug targets, suggesting their potential in drug development.

Saliva and Serum Cytokine Profiles During Oral Ulceration in Behcet's Disease

Tanya Novak et al.

Summary: Behcet's disease (BD) is characterized by high levels of cytokines in saliva, especially in patients with oral ulcers. Oral ulcers in BD may indicate reactivation of systemic inflammation, and monitoring cytokine levels in saliva could provide insights into disease activity and progression.

Genetic determinants of blood-cell traits influence susceptibility to childhood acute lymphoblastic leukemia

Linda Kachuri et al.

Summary: Through a genome-wide association study, blood-cell traits associated with childhood acute lymphoblastic leukemia susceptibility were identified. The study revealed that a genetically induced shift towards higher lymphocyte counts increases the susceptibility to ALL, while other cell types have opposite effects.

Advancing the use of genome-wide association studies for drug repurposing

William R. Reay et al.

Summary: Genome-wide association studies have provided important biological insights into complex diseases, offering opportunities for drug repurposing. Leveraging common variant genetics through approaches such as single-loci mapping to drug targets, transcriptome-wide association studies, gene-set association, causal inference by Mendelian randomization, and polygenic scoring, researchers aim to identify new treatment options more efficiently.
