Nanocatalysis meets microfluidics: A powerful platform for sensitive bioanalysis

Yanfeng Gao et al.

Summary: Accurate, sensitive, and highly selective detection of biological targets is crucial in various fields. The combination of nanocatalysis and microfluidics has shown great success in bioanalysis and offers numerous applications for different biological targets. This review summarizes the recent developments in nanocatalysis-assisted bioanalytical microfluidic devices, including various catalytically active nanomaterials, microfluidic approaches, detection techniques, and their applications. The integration of nanocatalysts for signal conversion and amplification is emphasized to enhance detection sensitivity. Furthermore, the challenges and future trends in nanocatalysis-assisted microfluidics are discussed, aiming to guide the development of practical point-of-care testing devices.

Phage-Based Profiling of Rare Single Cells Using Nanoparticle-Directed Capture

Yuan Ma et al.

Summary: The study demonstrates an integrated microfluidic chip using magnetic nanoparticles to efficiently capture single tumor cells, enable on-chip incubation, and facilitate in situ cell-surface protein expression analysis. By monitoring changes in the expression of multiple surface markers in response to CTC adherence, the researchers found fluctuations in the expression of Frizzled2 (FZD2) that reflected the microenvironment of the single cells. This platform has the potential to simultaneously screen multiple surface antigens in rare cells with single-cell resolution, providing further insights into biological heterogeneity and human disease.

Hexokinase 2 discerns a novel circulating tumor cell population associated with poor prognosis in lung cancer patients

Liu Yang et al.

Summary: This study demonstrates hexokinase-2 (HK2) as a metabolic function-associated marker for the detection of circulating tumor cells (CTCs) in non-small cell lung cancer (NSCLC) patients. The HK2(high)/CKneg CTCs exhibit metastasis and EGFR inhibitor resistance-related molecular signatures, and are selectively enriched in patients with EGFR(L858R) driver oncogene mutation. Patients with a larger number or proportion of HK2(high)/CKneg CTCs in the blood show poor therapy response and shorter progression-free survival.

Ultra-high-throughput single-cell RNA sequencing and perturbation screening with combinatorial fluidic indexing

Paul Datlinger et al.

Summary: The development of 'single-cell combinatorial fluidic indexing' (scifi) allows for cost-effective single-cell sequencing on a massive scale, combining preindexing of entire transcriptomes with single-cell RNA-seq using microfluidics. This approach significantly increases throughput and enables sample multiplexing in a straightforward manner.
