Asymmetric Catalytic Ring-Expansion of 3-Methyleneazetidines with a-Diazo Pyrazoamides towards Proline-Derivatives

Chiral nickel(II) complex catalyzed asymmetric (3+2) cycloaddition of α-diazo pyrazoleamides with 2-siloxy-1-alkenes

Summary: A highly efficient asymmetric (3 + 2) cycloaddition of alpha-diazo pyrazoleamides with silyl enol ethers was achieved using a chiral N,N'-dioxide-Ni(II) complex catalyst. The reaction involves the formation of a chiral nickel carbenoid intermediate and subsequent enantioselective cycloaddition. The desired dihydrofuran O,O-acetal derivatives were obtained in good yields (up to 90%) and high enantioselectivity (up to 99% ee) under mild reaction conditions and short reaction time. A possible transition state model was proposed based on the determination of the catalyst structure. Enantioselective Catalytic Hantzsch Dihydropyridine Synthesis

Summary: Optically enriched Hantzsch dihydropyridines with different ester groups were obtained through asymmetric catalytic cascade reaction. Efficient N,N'-dioxide/NiII or NdIII complex catalysts yielded a variety of products including drugs like nitrendipine, nimodipine, and felodipine, in high yields (up to 99% yield) with excellent enantioselectivities (up to 99% ee). Two enantiomers of the products could be readily obtained via ester group exchange, and axial chiral 4-arylpyridines were also formed through oxidation.

Enantioselective Photochemical Carbene Insertion into C-C and C-H Bonds of 1,3-Diketones by a Guanidine-Amide Organocatalyst

Summary: A visible-light-induced enantioselective free carbene transfer reaction of alpha-diazoesters with 1,3-diketones is achieved using a chiral bifunctional guanidine-amide organocatalyst. Different steric hindrances of alpha-diazoesters allow for selective insertion of free carbene into the C-C or C-H bonds of 1,3-diketones. The reaction enables the synthesis of 1,4-diketones with a quaternary carbon chiral center or chiral 2-substituted-1,3-diketones in good enantioselectivities. A stepwise triplet free carbene coupling pathway is suggested based on control experiments and DFT calculation. The high enantioselectivities are proposed to originate from hydrogen-bond and electrostatic interactions of the catalyst.

Copper-Catalyzed Asymmetric Diyne Cyclization via [1,2]-Stevens-Type Rearrangement for the Synthesis of Chiral Chromeno[3,4-c]pyrroles

Summary: In this study, a copper-catalyzed asymmetric cascade cyclization/[1,2]-Stevens-type rearrangement was reported to efficiently assemble a variety of valuable chiral chromeno[3,4-c]pyrroles bearing a quaternary carbon stereocenter with generally moderate to good yields and excellent enantioselectivities (up to 99% ee). Notably, this protocol represents the first example of catalytic asymmetric [1,2]-Stevens-type rearrangement based on alkynes and the first asymmetric formal carbene insertion into the Si-O bond.

Independent gradient model based on Hirshfeld partition: A new method for visual study of interactions in chemical systems

Asymmetric Catalytic (2+1) Cycloaddition of Thioketones to Synthesize Tetrasubstituted Thiiranes

Biocatalytic One-Carbon Ring Expansion of Aziridines to Azetidines via a Highly Enantioselective [1,2]-Stevens Rearrangement

Summary: The study reports the enantioselective one-carbon ring expansion of aziridines to form azetidines through engineered carbene transferase enzymes. A variant of cytochrome P450BM3, P411-AzetS, showed unparalleled stereocontrol (99:1 er) in the [1,2]-Stevens rearrangement and could override the inherent reactivity of aziridinium ylides to perform this rearrangement. By controlling the reactive aziridinium ylide intermediates, these evolvable biocatalysts enable a transformation not achievable by other catalyst classes.

Enantioselective [1,2]-Stevens rearrangement of thiosulfonates to construct dithio-substituted quaternary carbon centers

The bis-sulfuration of alpha-diazocarbonyl compounds was developed by utilizing thiosulfonates as the sulfenylating agent. This atom-economic process provides an efficient route to access novel chiral dithioketal derivatives with high yields (up to 90%) and enantioselectivities (up to 96:4 er).
