Fluorescence lifetime-based pH mapping of tumors in vivo using genetically encoded sensor SypHerRed

Summary: Changes in intracellular pH reflect the metabolic states of cancer cells during tumor growth and dissemination. Monitoring pH is essential for understanding the metabolic mechanisms supporting cancer progression. Genetically encoded fluorescent pH sensors provide real-time tracking of pH in specific subcellular compartments, but their ratiometric readout is not suitable for thick samples ex vivo or tissues in vivo, including solid tumors. Fluorescence lifetime imaging (FLIM) offers a promising alternative to conventional fluorescence microscopy. This study presents a quantitative approach to map pH in cancer cells and tumors in vivo using the fluorescence lifetime of a genetically encoded pH sensor called SypHerRed. The study demonstrates the utility of SypHerRed in visualizing pH in cancer cell culture and mouse tumor xenografts using FLIM and macroscopy. For the first time, the absolute pH value is obtained for tumors in vivo using an optical technique. Additionally, the study shows the simultaneous detection of pH and the endogenous fluorescence of metabolic cofactor NADH, providing complementary insights into the metabolic aspects of cancer. The fluorescence lifetime-based readout and red-shifted spectra of SypHerRed make it a promising instrument for multiparameter in vivo imaging applications.

Mapping intracellular pH in tumors using amide and guanidyl CEST-MRI at 9.4 T

Summary: A novel method for absolute pH(i) mapping using endogenous CEST-MRI was introduced in this study, which compensates for various concomitant effects. The method was validated in porcine brain lysates and demonstrated applicability in vivo in tumor-bearing mice. This non-invasive approach allows for potential clinical studies on pH(i) changes in tumors as a biomarker for cancer diagnosis or treatment monitoring.

Total protein assay by PCA-based RGB-spectrum conversion methods with smartphone-acquired digital images

Summary: This study demonstrates a method for determining total protein concentrations in aqueous solutions using smartphone-captured color images. The proposed method converts RGB values to score values using principal component analysis and generates reproduced absorption spectra. The results of total protein assays using this method were consistent with those obtained using a conventional spectrophotometer, without the need for monochromating equipment.

Geometrical pH mapping of Microfluids by principal-componentanalysis-based xyz-spectrum conversion method

Summary: The absorption spectra of bromothymol blue (BTB) solution in microfluidic devices were analyzed using PCA-based methods to map the pH values of fluids accurately. PDMS microfluidic devices were used to introduce solutions of pH indicators under different conditions, and RGB values were analyzed to convert into spectral data. The method showed high reproducibility and applicability in measuring pH gradients in microfluidic channels, providing spectrophotometric information in milliseconds without the need for monochromating equipment.
