Mapping enzyme catalysis with metabolic biosensing

Summary: Enzymes have diverse activities that surpass chemical catalysts, but engineering them for novel or enhanced activity is hampered by limited technologies for sensing product formation. This study presents a general and scalable approach that uses host cell metabolism as a biosensor to infer product formation, enabling the discovery of unexpected products and mapping the effects of mutagenesis through mass spectrometry. Additionally, a screening method combining microfluidics and mass spectrometry is reported for mapping catalysis, characterizing product range, and recovering sequences of variants with desired activities.

Continuous and automated slug flow nanoextraction for rapid partition coefficient measurement

Summary: An automated system for rapid determination of log K-ow for an array of compounds using slug flow nanoextraction (SFNE) enabled by a microfluidic chip is reported in this study. The method includes segmenting the injected volume into a series of slugs, compartmentalizing each octanol-water phase pair using an immiscible fluorous carrier fluid, and utilizing an online UV absorbance detector for concentration analysis to measure log K-ow efficiently. This approach allows for online calibration and can be completed in under 2 hours for a panel of seven pharmaceutical compounds at three different pHs.
