Multi-CRAFTI: Relative Collision Cross Sections from Fourier Transform Ion Cyclotron Resonance Mass Spectrometric Line Width Measurements

Summary: Determining collision cross sections (CCS) using the CRAFTI technique is limited by accurate pressure requirements in the mass spectrometer. The multi-CRAFTI approach allows for measurement of relative CCS without pressure knowledge and determination of absolute CCS using internal standards. Multi-CRAFTI generally yields smaller CCS compared to IMS, and computed structure CCS are smaller than experimental CCS as m/z increases. Relative CCS reach constant values at lower collision energies compared to absolute CCS when ions undergo similar collisional processes.

Frequency chasing of individual megadalton ions in an Orbitrap analyser improves precision of analysis in single-molecule mass spectrometry

Summary: By investigating the behavior of macromolecular single ions within the Orbitrap analyzer, this study aimed to enhance the performance of charge-detection mass spectrometry. The results showed that ions with a mass beyond one megadalton can be successfully trapped for seconds, enabling precise mass analysis with a resolution of over 100,000. It was also found that the frequency drift of single ions due to desolvation and charge stripping can be corrected, resulting in an improved ion sampling efficiency and mass precision and resolution.

Separation and Collision Cross Section Measurements of Protein Complexes Afforded by a Modular Drift Tube Coupled to an Orbitrap Mass Spectrometer

Summary: New developments in analytical technologies and biophysical methods have advanced the characterization of increasingly complex biomolecular assemblies. Ion mobility methods, especially, provide a new dimension of structural information and protein analysis based on collision cross sections (CCSs). This article presents a modular drift tube design capable of separating native-like proteins with high resolution, and combines it with high-resolution Orbitrap MS for complex analysis. Additionally, the ability to rapidly determine accurate CCSs for proteins without the need for calibrants or correction factors is demonstrated.

Symmetry of 4-Oxalocrotonate Tautomerase Trimers Influences Unfolding and Fragmentation in the Gas Phase

Summary: The recent discovery of asymmetric trimers in the tautomerase superfamily (TSF) adds structural diversity and reveals the higher stability of asymmetric trimers compared to symmetric ones. Utilizing native mass spectrometry and ultraviolet photodissociation allows for rapid and sensitive differentiation of symmetric and asymmetric trimers. The increased stability of asymmetric trimers may justify their evolution and concomitant diversification of the TSF, aiding in delineating the evolutionary history of the TSF.

Nanohydrophobic Interaction Chromatography Coupled to Ultraviolet Photodissociation Mass Spectrometry for the Analysis of Intact Proteins in Low Charge States

Summary: The direct correlation between proteoforms and biological phenotype calls for the exploration of mass spectrometry (MS)-based methods suitable for proteoform detection and characterization. In this study, nano-hydrophobic interaction chromatography (nano-HIC) is combined with ultraviolet photodissociation MS (UVPD-MS) to separate and characterize intact proteins and proteoforms. The method achieves high linearity, sensitivity, and sequence coverage for various proteins. Investigation of intact protein collisional cross sections during nano-HIC reveals semi-folded conformations in low charge states, enabling a different dimension of separation compared to traditional fully denaturing reversed-phase separations. The method is demonstrated using a mixture of intact proteins from Escherichia coli ribosomes, achieving high sequence coverage for various modified and unmodified proteoforms.

Advancing Orbitrap Measurements of Collision Cross Sections to Multiple Species for Broad Applications

Summary: Measurement of collision cross section (CCS) is a parameter reflecting the size and shape of an ion and provides structural information beyond molecular mass alone. Previously, isolation of individual charge states was required for CCS analysis, limiting throughput. In this study, CCSs were determined from commonly available mass spectra files using an Orbitrap mass spectrometer, allowing for simultaneous estimation of CCS for multiple charge states and expanding the applicability of this method.
