Detection of SARS-CoV-2 RNA Using a DNA Aptamer Mimic of GreenFluorescent Protein

Bria S. VarnBuhler et al.

Summary: This article describes a simplified method for RNA detection using simple unmodified DNA oligonucleotides, which can rapidly detect the target RNA's fluorescent signal. By developing a fluorogenic DNA aptamer called Lettuce, the researchers have achieved rapid detection of the target RNA by binding and activating the fluorescence of DFHBI-1T, a nonfluorescent molecule that resembles the chromophore found in green fluorescent protein.

Development of a novel PROTAC using the nucleic acid aptamer as a targeting ligand for tumor selective degradation of nucleolin

Lin Zhang et al.

Summary: In this study, a novel PROTAC ZL216 was developed by using nucleic acid aptamer AS1411 as a targeting ligand of nucleolin and conjugating it with a small molecule ligand of E3 ligase VHL, achieving selective degradation of breast cancer cells. This study broadens the methods of constructing PROTACs and provides a promising strategy for tumor-selective therapy.

Endogenous Copper for Nanocatalytic Oxidative Damage and Self-Protection Pathway Breakage of Cancer

Yuedong Guo et al.

Summary: This study focuses on using endogenous copper ions as a source of Fenton-like agents in cancer treatment, to disrupt the intrinsic protection pathway of cancer cells and induce apoptosis through catalytic oxidative damage reactions. The nanocatalyst shows excellent biosafety and enhanced therapeutic efficacy by simultaneously targeting cancer cell self-protection mechanisms and inducing oxidative damage.

Structurally defined polythiophene-based nanoprobe for selective Cu2+ detection in living cells

Fei Chen et al.

Summary: SDBCPs have unique photophysical and self-assembly properties, but their potential for sensing applications has been rarely explored. The novel SDBCP P3EPT(17)-b-P3ImHT(33) can self-assemble into nanofibers in DMSO and quasispherical micelles in water, and it has been utilized as a fluorescent nanoprobe for the selective and sensitive detection of Cu2+ with a detection limit as low as 0.13 nM. Moreover, fluorescence imaging studies have shown that PT-CPNs can effectively detect Cu2+ in living HeLa cells.

Copper Promotes Tumorigenesis by Activating the PDK1-AKT Oncogenic Pathway in a Copper Transporter 1 Dependent Manner

Jianping Guo et al.

Summary: Copper activates the PI3K-AKT oncogenic signaling pathway to promote tumorigenesis, while CTR1 is abnormally elevated in breast cancer and regulated by Nedd4l-mediated ubiquitination. Targeting the CTR1-copper node may have therapeutic relevance for hyperactive AKT-driven cancers.
